SNRPA1,an important splicing factor of the spliceosome,has been reported to be highly expressed in colorectal and hepatocellular carcinomas,but its expression and related mechanism of action in gastric cancer are not yet known.In this study,we screened SNRPA1 based on the differential gene expression profile of gastric cancer and high-throughput sequencing results.Through a series of bioinformatics analysis,such as expression level,transcriptional and mutational analysis,interaction network,and clinical baseline data analysis,we suggested that SNRPA1 showed high expression in various tumors including gastric cancer.By constructing SNRPA1 lentivirus in the gastric cancer cell line AGS,cell biology experiments such as CCK8 and clone formation were used to demonstrate that interfering with SNRPA1 could inhibit the growth proliferation and colony formation ability of human gastric cancer cells AGS.Further,to investigate the biological role of SNRPA1 in gastric carcinogenesis and development,transcriptomic sequencing was used to analyze the differential gene expression patterns before and after SNRPA1 interference,and it was found that genes such as CPA4,CST4,and NPPB were highly expressed in AGS cells after SNRPA1 interference while MKI67,PRKDC,CDK6,SMG1,MDN1 SNRPA1,MCM8,and other genes were lowly expressed.Pathway enrichment analysis revealed that the cell cycle,Fox O signaling pathway,TGF-β signaling pathway,and Erb B signaling were altered in AGS cells after SNRPA1 interference.This provides a good basis for further exploration of the development and mechanism of SNRPA1 on gastric cancer,and SNRPA1 may become a novel and specific biomarker for the treatment of gastric cancer.
