| Objective: It was found that Nesfatin-1 can improve obesity,and there are few studies on other mechanisms in this process except inhibition of feeding and improvement of lipid metabolism.Studies have found that obesity is essentially a chronic inflammatory state,and this state is the main cause of various metabolic syndromes.In order to explore the effect and mechanism of Nesfatin-1 on chronic inflammatory state in obese rats,this study established an obese rat model to observe the level of inflammatory factors regulated by Nesfatin-1 and analyze the effect of Nesfatin-1 on the expression level of inflammation-related miRNA.To explore the mechanism of Nesfatin-1 in the inflammatory process of obesity.Methods: The obese rats were randomly divided into 5 groups: blank control group,normal saline group,Nesfatin-1 low-dose group,Nesfatin-1 medium-dose group and Nesfatin-1 high-dose group.Rats in each group were injected with normal saline and Nesfatin-1 at different concentrations daily through tail vein for 5 weeks,during which the body weight and food intake of rats were recorded.Then the liver and adipose tissue of rats were weighed and pathological sections were made to observe the morphological changes of the liver and adipose tissue of rats.The expression levels of TNF-α and IL-10 in serum of rats were detected by Elisa to evaluate the improvement of inflammatory response in obese rats by Nesfatin-1.Fluorescence quantitative PCR(q PCR)was used to detect the expression of inflammation-related micro-ribonucleacid(miRNAs,miR)in the liver of rats,and the mechanism of Nesfatin-1 on improving the miRNA of the inflammatory response of rats was analyzed.Results :(1)Nesfatin-1 can improve body weight,food intake and liver weight of obese rats to a certain extent.According to the results of body weight gain of rats,the effect of Nesfatin-1 high-dose group was better than that of normal saline group(P < 0.05).According to the results of food intake in rats,the Nesfatin-1 low-dose group was better in inhibiting food intake.According to the comparison of liver weight,there was no significant difference between Nesfatin-1 dose groups and normal saline group.(2)HE staining results showed that,compared with normal saline group,Nesfatin-1 dose groups could improve the swelling of adipose tissue and reduce the steatosis in liver tissue.(3)Elisa results showed that compared with normal saline group,the expression of TNF-α in serum and the expression of IL-10 in Nesfatin-1 dose groups were decreased with significant difference(P < 0.05),but there was no significant difference among the three dose groups.(4)q PCR results showed that Nesfatin-1 reduced the expression level of miR155 and increased the expression level of miR146 a compared with normal saline group,with significant differences(P < 0.05).Conclusions :(1)Nesfatin-1 can improve food intake and reduce body weight and liver weight in obese rats.(2)Nesfatin-1 can alleviate the inflammatory response in obese rats.(3)Nesfatin-1 regulates inflammation by increasing Mir-155 and decreasing mi-146 a expression levels.These results suggest that Nesfatin-1 plays a regulatory role in the inflammatory process of obesity,which will have potential application in the treatment of metabolic diseases. |
