| Background and Objective:Acute lung injury/Acute respiratory distress syndrome(ALI/ARDS)is an inflammatory lung disease.It could cause high mortality rate due to various reasons.Although there have been many studies on the genomics,proteomics and related signal pathways of the disease,there is no clear study to show the most important genes acted in the disease.The purpose of this study is to explore the key genes with the strongest correlation with ALI/ARDS in the existing studies,The transcription factors regulating the key genes were predicted by text mining and bioinformatics analysis.Methods:1.All genes that related to ALI and ARDS are found through Coremine Medical,and then the interaction between genes and diseases was explored through Chilibot,a text mining network platform2.All genes that related to ALI and ARDS were analyzed for gene function and signal pathway,and,the protein interaction network was established by using String.The hub genes were screened by using the visualization tool Cytoscape.In addition,the transcription factor network was predicted by using networkanalyst network platform.3.The cell model of ALI/ARDS was established.The expression levels of transcription factors and hub gene were detected by reverse transcription quantitative polymerase chain reaction(RT-qPCR),and,Western blot(WB).Result:1.A total of 1247 genes related to ALI.and 2036 genes related to ARDS were found by text mining.After removing the duplicates,there were 1468 genes related to ALI/ARDS.2.Related genes are mainly enriched in the structure and function related to cytokines and chemokines.3.The Top ten hub genes in the protein interaction network:CXCL8,CXCL10,CXCL1,CXCR4,CCL5,CCR2,CXCL12,CXCL9,CC14 and CXCR34.The Top two in the transcription factor network are wrnip1 and CBFB.5.The results of RT qPCR and WB showed that the expression of transcription factor CBFB and hub gene CCR2 in LPS treated group were significantly higher than that in normal group.Conclusion:By using the methods of medical text mining and bioinformatics,we found the hub genes and transcription factors related to acute lung injury/acute respiratory distress syndrome,and verified the expression difference of transcription factor CBFB and hub gene CCR2 in the disease model.According to cell experiment,this study would provide a new idea for the treatment of acute lung injury/acute respiratory distress syndrome in the future. |
