Objectives:In order to investigate the effect of intestinal endocrine hormone changes on hypertension,the expression changes of toll like receptor 4(TLR4),vasoactive intestinal peptide(VIP),glucagon like peptide-2(GLP-2)and glucagon like peptide-2receptor(GLP-2R)in the intestinal tissues of hypertensive rats and gentamicin-treated rats were detected in this study.Methods:Rats were fed high fat diet to establish the model of hypertension rat.Eight-week-old male Sprague-Dawley rats were randomly divided into three groups after one week of adaptive feeding:control group[(Provide normal-diet),n=11],hypertension group[(Provide high-fat diet,n=10)]and gentamicin group[(Provide high-fat diet),n=8)].The blood pressure levels of 3 groups were measured noninvastively from the tail artery asbaseline levels,The blood pressure levels of rats were monitored every week from the 5th week until the hypertension level was reached in the high-fat feeding group.From the 9th week,the gentamicin group rats was intragastrically administered with gentamicin sulfate,and the control group and the hypertension group were intragastrically administered with normal saline.At the end of the 13th week,rats in each group were sacrificed,and intestinal tissues ileum and colon were collected.The reverse transcription levels of TLR4,VIP,GLP-2 and GLP-2R mRNA in ileum and colon tissues of rats in the three groups were detected by real-time q-PCR.Immunohistochemical staining was used to detect the expression of TLR4,VIP,GLP-2 and GLP-2R proteins in ileum and colon tissues of rats in three groups.Results:1.The level of TLR4 mRNA in ileum and colon of the hypertensive group was significantly higher than that in the control group(P<0.05),and the level of TLR4 mRNA in ileum and colon of the gentamicin group was significantly lower than that in the hypertensive group(P<0.05)and the control group(P<0.05).The expression of TLR4 protein in colon of the hypertensive group was significantly higher than that in the control group(P<0.05),and the expression of TLR4 protein in colon of the gentamicin group was significantly lower than that of the hypertensive group(P<0.05),but there was no significant difference between the gentamicin group and control group(P>0.05).And the expression of TLR4 protein in ileum showed no significant difference among the three groups(P>0.05).2.The level of GLP-2 mRNA in ileum and colon of the hypertensive group was significantly lower than that in the control group(P<0.05),and the level of GLP-2 mRNA in the gentamicin group was significantly higher than that in the hypertensive group(P<0.05)and significantly lower than that in the control group(P<0.05)in ileum and colon.The expression of GLP-2 protein in ileum and colon in the hypertension group was significantly lower than that in the control group(P<0.05).The expression of GLP-2 protein in ileum and colon in the gentamicin group was significantly higher than that in the hypertension group(P<0.05),there was no significant difference between the gentamicin group and the control group(P>0.05).3.The level of GLP-2R mRNA in ileum and colon of the hypertensive group was significantly higher than that in the control group(P<0.05).The level of GLP-2R mRNA in colon of the gentamicin group was significantly lower than that in the hypertension group(P<0.05)and the control group(P<0.05).The level of GLP-2R mRNA in ileum of the gentamicin group was significantly lower than that in the hypertension group(P<0.05),and there was no significant difference between the gentamicin group and the control group(P>0.05).The expression of GLP-2R protein in ileum and colon of the hypertensive was significantly higher than that in the control group(P<0.05).The expression of GLP-2R protein in ileum of the gentamicin group was significantly lower than that in the hypertension group(P<0.05)and significantly higher the control group(<0.05).The expression of GLP-2R protein in colon of the gentamicin group was significantly lower than that in the hypertensive group(P<0.05),but there was no significant difference between the gentamicin group and the control group(P>0.05).4.The level of VIP mRNA in ileum of the hypertensive group was significantly lower than that in the control group(P<0.05),and that in the gentamicin group was significantly higher than that in the hypertensive group(P<0.05),and was significantly lower than that in the control group(P<0.05).There was no significant difference in VIP mRNA level in colon between the hypertension group and the control group(P>0.05),The level of VIP mRNA in colon of the gentamicin group was significantly higher than that in the control group(P<0.05),and the hypertension group(P<0.05).The expression of VIP protein in ileum of the hypertensive group was significantly lower than that in control group(P<0.05),and the expression of VIP protein in the gentamicin group was significantly higher than that in the hypertensive group(P<0.05),but there was no significant difference between the gentamicin group and control group(P>0.05).There was no significant difference in VIP protein expression in colon tissues of rats in the three groups(P>0.05).Conclusion:In the hypertensive group,the expression of intestinal TLR4 was up-regulated,leading to intestinal inflammation and decreased intestinal VIP expression.The expression of GLP-2 was down-regulated,and the expression of GLP-2Rwasup-regulated.Gentamicin sulfate may inhibit TLR4 pathway,decrease the production of inflammatory factors,relieve intestinal inflammation,increase GLP-2 and VIP expression,protect intestinal mucosal barrier,thus alleviate the hypertension induced by high-fat diet in rats.
