Objective:To study the relationship between the expression level of Quiescin Sulfyl Oxidase 1(QSOX1)and clinicopathological features and prognosis of Pancreatic Ductal Adenocarcinoma(PDAC).Methods:1.The clinicopathological data of 69 patients with PD AC who underwent "Radical resection of pancreatic cancer" in Nanchong Central Hospital from August 2014 to August 2019 were retrospectively analyzed.The paraffin sections of cancer and paracancer tissues,21 benign pancreatic disease tissues and 8 normal pancreatic tissues were collected for immunohistochemical staining.The expression of QSOX1 in the above tissues and its relationship with the clinicopathological features of PDAC were analyzed.2.The accuracy of QSOX1 in diagnosing PDAC was analyzed by SPSS software;3.Kaplan-meier survival curve and Cox regression model were used to analyze the relationship between QSOX1 expression level and survival prognosis of PDAC patients.Results:Among the 69 PDAC paraffin sections,58 samples showed high expression of QSOX1,and the expression level of QSOX1 in PDAC tissues was higher than that in paracancerous tissues(Z=-5.537,P<0.001),benign pancreatic disease tissues(Z=-4.913,P<0.001),and normal pancreas tissues(Z=-3.850,P<0.001),the difference was statistically significant;The expression level of QSOX1 in poorly differentiated PDAC was higher than that in moderately differentiated PDAC(Z=-2.042,P=0.041)and highly differentiated PDAC(Z=-2.611,P=0.009);The area under ROC curve of QSOX1 for the diagnosis of PDAC was 0.856,with 84.1%sensitivity,75.9%specificity,89.2%positive predictive value,66.7%negative predictive value and 81.6%coincidence rate;QSOX1 was associated with preoperative CA19-9 level(Z=-4.887,P<0.001),lymph node metastasis(Z=-3.147,P=0.002),vascular invasion(Z=-2.516,P=0.012),and nerve invasion(Z=-2.340,P=0.019);Median survival time of patients in the high QSOX1 group was shorter than that in the low QSOX1 group(8/15 months,χ2=4.909,P=0.027).Cox univariate regression analysis indicated lymph node metastasis(HR=0.349,95%CI=0.187-0.652,P=0.001),preoperative CA19-9 level(HR=0.208,95%CI=0.074-0.588,P=0.003)and QSOX1 expression levels(HR=0.512,95%CI=0.273-0.962,P=0.037)were correlated with prognosis of PDAC patients.Cox multivariate regression analysis indicated lymph node metastasis(HR=0.375,95%CI=0.180-0.78,P=0.009),preoperative CA19-9>37.0IU/mL(HR=0.145,95%CI=0.045-0.460,P=0.001)and high QSOX1 expression(HR=0.443,95%CI=0.226-0.868,P=0.018)were independent risk factors for poor prognosis in PDAC patients.Conclusion:QSOX1 was highly expressed in PDAC,and was significantly correlated with PDAC differentiation,vascular and nerve invasion,and lymph node metastasis.High QSOX1 expression is an independent risk factor for poor prognosis in PDAC patients,predicting a shorter postoperative survival.
