Objective:The abnormal expression of coding genes and non coding genes is closely related to the occurrence and development of tumors.In recent years,with the gradual deepening of the research on the genetics and epigenetics of glioblastoma,many coding genes have been gradually applied to the development of therapeutic drugs.This paper mainly explores the role of FAM120A and its protein expression in the occurrence and development of glioblastoma and its mechanism.Methods:Through biological information analysis,the search results in CGGA database showed that the expression of FAM120A in glioblastoma was higher than that in normal brain tissue.Then,the expression of FAM120A in cells and tissues was evaluated in vitro.The cell lines with the most significantly increased expression were screened,and then lentivirus was transfected with scrambled siRNA,FAM120A-siRNA and PI3K/AKT signal pathway inhibitor(LY294002).The experimental results were compared with the control group to evaluate the ability of cell proliferation,invasion and migration,and the activation of PI3K/AKTsignaling pathway.Finally,subcutaneous tumorigenesis was performed in nude mice to verify the relationship between FAM120A and PI3K/AKT signaling pathwayResults:Theexpression of FAM120A was increased in glioblastoma specimens.In vitro and in vivo experiments,after interfering with the expression of FAM120A,the related factors of PI3K/AKT signal pathway(Bcl-2,p-AKT(ser473),p-AKT(thr308))decreased,while the expression of Bax increased.After inhibiting the expression of FAM120A,the proliferation,invasion and migration of tumor cells decreased.Conclusion:The abnormal expression of FAM120A may regulate the proliferation,invasion and migration of glioblastoma by activating PI3K/AKT signaling pathway.This study is expected to provide a new marker for molecular targeted therapy of glioblastoma and a new research direction for improving the prognosis of patients.
