Multiple myeloma is a serious malignancy derived from plasma cells,accounting for 1% of all cancers and the second most common hematologic malignancy.Common clinical symptoms include anemia,bone destruction and renal insufficiency.In recent years,the treatment of multiple myeloma has made great progress,but it is still an incurable blood tumor disease due to its high recurrence rate.Therefore,it is necessary to find new drugs with lower toxicity and more effective.Morusin is the main phenolic component in the root bark of morusin,which has attracted the attention of related researchers due to its biological activity and good antioxidant capacity,and has been found to have good anti-cancer,anti-inflammatory and anti-aging effects.At present,there are no relevant reports about the effects of sansin on multiple myeloma cells.Therefore,we further explore the effects and mechanism of sansin on multiple myeloma cells,which will provide some experimental theoretical basis for the treatment of multiple myeloma patients and the development of corresponding anti-cancer drugs.The main results:(1)The showed that morusin could inhibit the proliferation of ARP-1 cells and induce the apoptosis of ARP-1 cells in a time-dependent and concentration-dependent manner.(2)Sequencing results showed that the effect of morusin on ARP-1 cells could inhibit the proliferation and promote apoptosis of ARP-1cells by regulating related genes in PI3K/AKT signaling pathway.(3)Western Blot showed that the expression levels of Bax,Caspase-3 and PTEN were significantly up-regulated at the protein level,while the expression levels of BCL-2,P-PI3K and P-AKT1 were significantly down-regulated.(4)The total expression levels of PI3K and AKT were not significantly changed.RT-qPCR results showed that the expression levels of Bax,Caspase-3 and PTEN were significantly up-regulated at RNA level,while the expression levels of BCL-2,PI3K and AKT1 were significantly down-regulated.In conclusion,morusin can inhibit the proliferation and apoptosis of ARP-1 cells.Through mechanism studies,it was found that the inhibition of morusin on the growth and apoptosis of ARP-1 cells may be related to PI3K/AKT signaling pathway.
