Objective:By collecting and collating the medical records of Professor Lv Ying’s treatment of autism spectrum disorder,this thesis analyzes the clinical characteristics and curative effect of the disease,discusses the feasibility and practicability of "Yi Tu Zai Mu" method for clinical guidance and summarizes the clinical experience.The theis also uses network pharmacology and molecular docking technology to explore the effective ingredients,potential targets and mechanism of action of representative prescriptions,which provides the modern scientific basis for the treatment of autism spectrum disorders by "Yi Tu Zai Mu".Methods:The thesis collects medical records of patients with autism spectrum disorder from January 2020 to January 2022 for retrospective analysis of general conditions,information of four diagnoses and case characteristics.It extracts the representative prescription of "Yi Tu Zai Mu" through factor analysis,and also combines with literature research related to mu,tu in the past dynasties,tracing the source,essence and development extension of those,clarifying the connotation of "Yi Tu Zai Mu" method,summarizing Professor Lv Ying’s experience in treating autism spectrum disorder with "Yi Tu Zai Mu" method.By analyzing representative prescriptions with network pharmacology research and Efficacy assessment,the molecular docking verification on the key components and the key targets were obtained.Result:1.This research collects 98 patients with autism spectrum disorder,with a total of 296 diagnose times,including 85 male patients and 13 female patients.The onset age was mainly in two to four years old,and the core symptoms were mainly language disorders,followed by social disorders and abnormal behaviors.The common accompanying symptoms included mood disorders,movement disorders,sleep disorders,food intolerance,partial eating,constipation,loose stool,irregular urination,etc.The tongue and pulse were mainly manifested as light red tongue,thin and whitish tongue coating,and thin pulse.A total of 16 traditional Chinese medicines were used in "Yi Tu Zai Mu" method.Factor analysis suggested that the main prescription for treating deficiency was Shaoyao Gancao decoction and the total effective rate of treatment was 82.5%.2.The results of network pharmacology research showed that the main effective components of Shaoyao Gancao decoction were quercetin,kaempferol,18βglycyrrhetinic acid,phytosterol,medicarpin,etc.The core targets were RELA,TNF,STAT3,MYC,AKT1,FOS,NR3C1,ESR1,etc.Gene set enrichment analysis of KEGG signaling pathway mainly involved JAK-STAT signaling pathway and adipokine signaling pathway.Molecular docking results showed that the binding energy of quercetin,kaempferol and 18β glycyrtinic acid with RELA,TNF and STAT3 were low,and the binding outcome was good.Conclusions:Shaoyao Gancao decoction has a definite effect on the treatment of autism spectrum disorder and can improve the patient’s language ability,social ability and abnormal behavior.It also can improve the mood,exercise,sleep,diet,urine,stool obviously.With multi-component,multi-targets and multi-pathway,the mechanism of Shaoyao Gancao decoction in the treatment of autism spectrum disorder using neuroprotective effects such as inhibiting neuroinflammation,reducing neurotoxicity and promoting nerve cell proliferation to improve the symptoms of patients with autism spectrum disorder.Therefore,the "Yi Tu Zai Mu" method can help patients recover,enhance vitality and improve the condition.
