| Objective:This study aimed to explore the correlation between aldehyde dehydrogenase 2(ALDH2)rs671 gene polymorphisms and acute ischemic stroke(AIS)occurrence,cerebral blood flow(CBF)on the affected side and prognosis,which provided a new basis for the prevention and treatment of high-risk groups of AIS.Methods:Thirty cases of AIS patients treated in Affiliated Hospital of Chifeng University were selected as the case group,and 30 cases of individuals without cerebrovascular disease during the same period were selected as the control group.All included subjects were collected from gender,age,history of smoking and alcohol consumption,homocysteine(HCY),total cholesterol(TC),fasting blood glucose,low density lipoprotein-cholesterol(LDL-C),triglyceride(TG),creatinine(Cr),white blood cells,hemoglobin,platelets and history of hypertension and diabetes.The DNA of the subjects was extracted,and the ALDH2 genotype of the two groups of subjects was obtained by Sanger sequencing.The CBF was examined under three-dimensional arterial spin labeling(3D-ASL)and processed by Functool software..The AIS subgroup was grouped according to the 90-day prognosis and ALDH2 genotype,and the general data of each group were statistically analyzed,and the correlation between ALDH2 genotype and AIS,prognosis and CBF on the affected side was analyzed.Results:1.Three genotypes of GG,GA and AA were detected at the ALDH2 rs671 site.The genotype and allele distribution conform to the Hardy-Weinberg equilibrium law,and the P values are 0.989 and 0.970,respectively.2.Compared with the control group,there were significant differences in diabetes history,hypertension history,HCY,TC,fasting blood glucose,TG,LDL-C and leukocytes in the AIS case group(P<0.05),there were statistical differences in the genotype distribution between the two groups(χ~2=0.593,P=0.032<0.05);and there were statistical differences in allele distribution between the two groups(χ~2=4.093,P=0.043<0.05).3.Compared with the group with good prognosis of AIS,there was a significant difference in the distribution of LDL-C and HCY(P<0.05),and the difference in the distribution of diabetes history between the two groups reached a statistically significant marginal level(P=0.066).The difference in genotype distribution between the two groups reached a marginally significant level(P=0.066).The difference in allele distribution between the two groups reached a marginally significant level(P=0.052),and the CBF in the group with a good prognosis was higher than that in the group with a poor prognosis,and the difference was statistically significant(t=3.80,P=0.001<0.05).4.Compared with the GA+AA group,there was a statistically significant difference between LDL-C and diabetes history in the AIS genotype GG group(P<0.05),and the CBF in the AIS genotype GG group was higher than that in the GA+AA group,and the difference was statistically significant(t=3.41,P=0.002<0.05).5.Regression analysis showed that fasting blood glucose(P=0.012,OR=2.734,95%CI1.250-5.983),LDL-C(P=0.017,OR=3.509,95%CI1.251-9.847)and HCY(P=0.038,OR=1.236,95%CI1.012-1.509)were independent risk factors for the occurrence of AIS.Conclusion:1、ALDH2 rs671 gene polymorphisms may be related to AIS susceptibi lity and prognosis,A allele may be a risk factor for occurrence and prognosis of AIS.2、ALDH2 rs671 gene polymorphisms may be related to CBF on the affected side of AIS patients.3、Fasting blood glucose,LDL-C and HCY may be independent risk factors for the occurrence of AIS. |
