| Objective: Based on SPSS Modeler 18.0 and SPSS Statistics 25.0software,data mining was conducted on the treatment of advanced cervical cancer by Dr.Chunzhuang Jiang,the chief physician,to summarize his medication rules and ideas,and to analyze the active ingredients,targets and signaling pathways of high-frequency drugs through pharmacological studies of Chinese medicine network,so as to provide empirical guidance for clinical treatment of advanced cervical cancer.Methods: In this study,a retrospective study was conducted to collect cases and prescriptions of advanced cervical cancer treated by Dr.Chunzhuang Jiang,the chief physician,from 01/01/2010 to 01/31/2023.Using Microsoft Excel 2010,SPSS Modeler 18.0 and SPSS Statistics 25.0software platform for data mining,we obtained the frequency of medicine use,the classification pattern of taste and nature,and efficacy of the treatment of advanced cervical cancer by Dr.Chunzhuang Jiang through frequency and frequency statistics,association rule analysis and cluster analysis,and other research methods.The data were then visualized and displayed.The active ingredients and targets of the core HF drugs were obtained from TCMSP database,and the targets of cervical cancer were obtained from GeneCards and other database platforms,and the intersecting targets were uploaded to String database to construct protein interactions;the Metascape platform was used to enrich the intersecting targets,and the main biological processes and metabolic pathways of the core HF drugs for the treatment of cervical cancer were derived.The network was constructed and visualized by Cytoscape 3.9.1software for the active ingredient-target and intersection target-pathway of the core HF drugs.In this way,we summarized the rules and ideas of drug use for advanced cervical cancer by Dr.Chunzhuang Jiang and further analyzed the mechanism of action of the core HF drugs.Results: 1.70 patients were enrolled in this study,with an average age of 54.8 years,and the most patients aged 51-60 years were 25 cases,accounting for 35.71% of the total.2.The highest frequency of identification and typing was for the damp-heat,stasis and toxicity type and the spleen-kidney yang deficiency type,both of which were 23 cases,accounting for 32.86% of the total.3.The top ten drugs in terms of frequency were: Astragalus,Atractylodes,Dendrobium,Tu Fu Ling,Paeonia,Xia Ku Cao,Rhizoma,Che Qian Zi,Gui Zhi and Jiang Han Xia.4.The frequency of sexual taste and meridian is sorted from high to low,medicinal properties: cold,warm,flat,cool,hot,medicinal taste:bitter,sweet,pungent,sour,salty,astringent,light,meridian: liver meridian,lung meridian,spleen meridian,stomach meridian,kidney meridian,etc.5.The drugs with greater than 5% frequency of efficacy categorization were deficiency tonics,heat-clearing drugs,diuretics and dampness-permeating drugs,and blood-stasis-activating drugs.6.High-frequency drugs were analyzed by association rules,and a total of5 high-frequency pairs of drugs and 23 high-frequency corner drug rules were obtained;clustering analysis yielded a total of 8 groups of core drug combinations.7.The pharmacological analysis of Chinese medicine network,obtained the core HF drugs,namely,20 active ingredients of Astragalus,7 of Atractylodes,15 of Dendrobium,15 of Tu Fu Ling,13 of Paeonia,and 64 active ingredients after de-weighting,the main active ingredients are quercetin,kaempferol,β-sitosterol,24-hydroxy-11-deoxyglycyrrhetinic acid,7-O-methyl-isomicroconvex sabinol,etc.;further obtained 438 active targets of Astragalus,20 of Atractylodes,90 of Dendrobium,306 of Tu Fu Ling,114 of Paeonia,and281 after de-weighting.20,Atractylodes macrocephala,90,Dendrobium,306,and 114 component action targets,and 281 targets were obtained after de-weighting,and 1527 cervical cancer-related targets were obtained,with 131 intersecting targets of both,of which MAPK3,MAPK1,AKT1,RAF1,RELA,etc.were predicted to be important targets for core HF drug treatment of cervical cancer.8.The KEGG signaling pathways of core HF drug therapy for cervical cancer are Pathways in cancer,Lipid and atherosclerosis,Hepatitis B,etc.The main biological processes involved include response to hormones,response to inorganic substances,and cellular response to lipid,etc.The target functions are mainly enriched in protein kinase activity,DNA-binding transcription factor binding,transcription factor binding,etc.Conclusion: 1.Dr.Chunzhuang Jiang,the chief physician of the treatment of advanced cervical cancer,used cold and warm drugs,bitter and sweet drugs,and the meridians are mainly liver,lung,spleen,stomach and kidney meridians,which are closely related to the 12 meridians.Therefore,the main treatment is to nourish the deficiencies of the liver,spleen and kidneys,and to take into account the five and six internal organs as a whole;the medicine used is to clear heat and dry dampness,strengthen the spleen and dispel dampness,invigorate blood circulation and remove blood stasis,taking into account the draining of the liver and Qi,and warming the interior and dispersing cold.2.Commonly used pairs of drugs are: ginger half Xia → Astragalus,wine Huangjing →Astragalus,Cornu Cervi Pantotrichum → Astragalus;commonly used corner medicine: Che Qian Zi + Atractylodes → Astragalus,ginger half Xia +Peony → Astragalus,ginger half Xia + Atractylodes → Astragalus,Coix Seed + Tu Fu Ling → Astragalus,Poria + Xia Gu Cao → Astragalus,Gui Zhi + Peony → Astragalus;commonly used drug combinations are: Fu Shen-Tian Ma-Zhi Mu-Chuan Xiong-Dan Shen-Huang Lian-light bamboo leaf-Scutellaria,Dendrobium-Xuan Shen,Chuan Niu Kne-Angelicae-Curcuma longa-Aromatic Herb,Paeonia lactiflora-Roasted Licorice-Ginger Semen,Plantago ovata-Coix Seed-Branched Atractylodes-Astragalus-Raw Atractylodes,Gui Zhi-Cumin-Tu Fu Ling-Yin Chen-Poria-Dry Ginger.3.The pharmacological research method of Chinese medicine network obtained 64 active ingredients of core HF drugs,131 targets are closely related to the treatment of cervical cancer,quercetin,kaempferol,β-sitosterol,24-hydroxy-11-deoxyglycyrrhetinic acid,7-O-methyl-isoconvex sabinol,etc.are important active ingredients,MAPK3,MAPK1,AKT1,RAF1,RELA,etc.are the main potential action MAPK3,MAPK1,AKT1,RAF1,RELA,etc.are the main potential targets,mainly involving Pathways in cancer,Lipid and atherosclerosis,Hepatitis B and other signaling pathways to regulate protein kinase activity,etc.,so as to achieve effective treatment of cervical cancer. |
