| Acanthopanax senticosus(Acanthopanax senticosus Rupr.et Maxim)is a plant of the pentaceae family,whose main medicinal parts are subterrane.In this paper,the adsorption performance of A.senticosus biochar was evaluated through the study of the effective extraction and extraction mechanism of A.senticosus and the pyrolysis of A.senticosus drug residue to prepare biochar.1.Establish an analytical method for the simultaneous determination of eleutheroside B,eleutheroside E,rutin,hypericin and Quercitrin content by high performance liquid spectrometry.Chromatographic conditions:Column:Ecosil HPLC C18column(250 mm×4.6mm,5μm);Mobile phase:0.2%aqueous phosphoric acid(A)-acetonitrile(B);Flow rate:1m L/min;Detection wavelength:275 nm;Column oven temperature:35℃,injection volume:10μL.Gradient elution conditions:0-8 min,8%(B);8-15 min,8-10%(B);15-25 min,10%(B);25-30 min,10-18%(B);30-45 min,18%(B);45-55 min,18-30%(B)。2.The process parameters of negative pressure cavitation-assisted deep eutectic solvent extraction of the active ingredients in A.senticosus were determined,and the effects of extraction pressure,extraction time and liquid-material ratio on the extraction of active ingredients from A.senticosus by negative pressure cavitation-assisted deep eutectic solvent were investigated by single factor screening,and the BBD design-response surface method was applied to investigate the effects of extraction pressure,extraction time and liquid-material ratio on the extraction of active ingredients from A.senticosus by negative pressure cavitation-assisted deep eutectic solvent.The optimal extraction process was determined as follows:extraction pressure:-0.07 Mpa,extraction time:25 min,liquid-solid ratio:20∶1(m L/g).Among them,the molar ratio composition of deep eutectic solvent is:choline chloride:lactic acid(Ch Cl-LA)=1∶3,water content 30%.Under the above optimal conditions,the contents of eleutheroside B,eleutheroside E,rutin,hypericin and quercitrin were 1.73 times that of the traditional solvent methanol extraction.3.Through quantum chemical calculation combined with molecular dynamics simulation,the dissolution behavior of eleutheroside B,eleutheroside E,rutin,hypericin and quercitrin in Ch Cl-LA solvent,solvent reachable surface area,hydrogen bond formation with solvent,and interaction energy with solvent were explored,and it was found that these five target compounds showed better dispersion state,accessible surface area,interaction energy and hydrogen bond life in Ch Cl-LA solvent system.At the same time,infrared spectroscopy,scanning electron microscopy and nuclear magnetic resonance hydrogen spectroscopy were used to further verify the efficient extraction of five target compounds from negative pressure cavitation-assisted Ch Cl-LA solvent,and the mechanism of efficient extraction of five target compounds by negative pressure cavitation-assisted Ch Cl-LA was explored.4.Biochar was prepared by A.senticosus drug residue,and the factors affecting the preparation of biochar were explored.By investigating the adsorption capacity of biochar on methylene blue,the optimal preparation process parameters were determined and the biochar was characterized.The adsorption capacity of A.senticosus drug residue biochar was investigated by the adsorption capacity of methylene blue,and the optimal conditions for the adsorption of methylene blue were as follows:liquid-material ratio of 6∶1(m L/g),carbonization temperature of 500°C,activation time of 4 h,under these conditions,the adsorption rate of methylene blue reached 39.48 mg/g.The experimental results of this study can provide a theoretical reference for the development and comprehensive utilization of active ingredient resources in Acanthopanax senticosus. |
