| Objective: Based on traditional Chinese medicine inheritance assistance platform software(V2.50),summarize and analyze the drug characteristics and prescription rules of traditional Chinese medicine in the treatment of allergic skin diseases,The efficacy test of the selected core prescription was conducted to observe its anti-allergic effect and mechanism on mice,so as to provide direction and basis for the clinical treatment of allergic skin diseases prescription.Methods:1.Relevant literatures on the treatment of allergic skin diseases with TCM in CNKI were summarized,and corresponding prescription database was screened and constructed.The frequency,efficacy and meridian distribution of TCM in the prescription for the treatment of allergic skin diseases were statistically analyzed with the help of the platform software of TCM inheritance,so as to discover the medication rules and obtain the new prescription.2.In the preliminary experiment,the core prescription with obvious anti-allergic effect was screened out,and 36 KM mice,half male and half female,were selected and divided into blank control group,model group,loratadine group,and core prescription high dose,medium dose and low dose groups with 6 mice in each group according to random number table method.After 3 days of adaptive feeding,the administration began.The blank control group and the model group were given normal saline intragastric administration,while the loratadine group and the core prescription group were given the corresponding drug intragastric administration every day.Mice in the above groups were given intragastric administration once a day for 7 days.One hour after the last administration,except for blank group,mice in other groups were injected with 0.0125% low molecular dextran solution through tail vein to induce pruritus.Blank group was replaced with normal saline.The experimental indexes were as follows: The degree of systemic skin pruritus induced by tail vein injection of low molecular dextran solution(scratching latency,scratching times and duration of scratching),and the inhibition rate was calculated.The contents of histamine(HIS),Ig E,IFN-γ and IL-4 in serum were detected by ELISA.Results:1.The frequency of medication and the rule of four qi and five tastes:495 prescriptions for the treatment of allergic skin diseases were screened,involving 297 Chinese herbs,and 60 of them appeared more than 20 times.Among them,the top 10 most frequently used TCM were Saposhnikovia divaricata(Trucz.)Schischk、Angelica Sinensis、Periostracum Cicadae、Cynanchum otophyllum Schneid、Rehmannia glutinosa(Gaertn.)Li-bosch、Nepeta cataria L、Cortex Dictamni,Astragalus membranaceus(Fisch.)Bunge 、 Atractylodes macrocephala Koidz 、Ligusticum chuanxiong Hort.The frequency of medicinal properties was mainly concentrated in cold,warm and flat,the taste of medicinal products was mainly concentrated in sweet,bitter and spicy,and the meridians of homing were mainly concentrated in liver meridian,spleen meridian and lung meridian.2.Combination rules:there were 33 commonly used TCM combinations(frequency≥60 times),such as Nepeta cataria L-Saposhnikovia divaricata(Trucz.)Schischk,Saposhnikovia divaricata(Trucz.)Schischk-Periostracum Cicadae,Angelica Sinensis-Saposhnikovia divaricata(Trucz.)Schischk,etc.Among them,the use frequency of Nepeta cataria L-Saposhnikovia divaricata(Trucz.)Schischk combination accounted for 22.4%,which was the most frequently used TCM combination,and the confidence of the combination rule of Nepeta cataria L-Saposhnikovia divaricata(Trucz.)Schischk was the highest,about0.83.3.Core combinations and new prescription rules: there were 28 core combinations in total.The top 5 combinations were Nepeta cataria L-Saposhnikovia divaricata(Trucz.)Schischk-Periostracum Cicadae,Nepeta cataria L-Arctium lappa L-Semen Sesami Nigrum,Fallopia multiflora(Thunb.)Harald-Astragalus membranaceus(Fisch.)Bunge-Tribulus ter restris L.,Fallopia multiflora(Thunb.)Harald-Ligusticum chuanxiong Hort-Tribulus ter restris L.,Orange peel-Atractylodes Lancea(Thunb.)DC.-Magnolia officinalis Rehd.et Wils.,and 14 new prescriptions were obtained,representedby "Nepetacataria LSaposhnikoviadivaricata(Trucz.)Schischk,Periostracum,Cicadae,Dark Plum Fruit,Stellaria dichotoma var.Lanceolata Bge.".4.In antiallergic experiments,compared with blankgroup,scratching times and scratching durationin model group were significantly increased(P<0.01),while scratching latency was shortened(P<0.05);In terms of scratching latency of mice,compared with model group,loratadine group could prolong scratching latency of mice(P<0.05),and core prescription high dose group could significantly prolong scratching latency of mice(P<0.01).In terms of scratching times of mice,compared with model group,scratching times of loratadine group and core prescription high dose and medium dose group were significantly decreased(P<0.01),while scratching times of core prescription low dose group were decreased(P<0.05);In terms of scratching duration,compared with model group,scratching times of loratadine group and core prescription high-dose,medium dose and low dose groups were significantly shortened(P<0.01).5.In the measurement of observed indicators,compared with blank group,The serum IL-4 content of model group was increased(P<0.05),The contents of histamine(HIS)and Ig E were significantly increased(P<0.01),and the content of IFN-γ was significantly decreased(P<0.01);In terms of histamine(HIS)content,compared with model group,The content of histamine(HIS)in serum of loratadine group and core prescription high dose,medium dose and low dose group was significantly decreased(P<0.01);In terms of Ig E content,compared with model group,Ig E content in serum of mice in low dose group of core prescription was decreased(P<0.05),and Ig E content in serum of mice in loratadine group and high and medium dose groups of core prescription was significantly decreased(P<0.01);In terms of IFN-γcontent,compared with model group,serum IFN-γ content in loratadine group and high and medium dose group of core prescription was significantly increased(P<0.01);In terms of IL-4 content,compared with model group,The serum IL-4 content in loratadine groupand high dose group of core prescription was significantly decreased(P<0.01),and the serum IL-4 contentin medium and low dose group of core prescription was decreased(P<0.05).Conclusion: 1.In The treatment of allergic skin diseases by TCM,The main medicines were strengthening body resistance and eliminating evil,dispelling wind and relieving pruritic,promoting blood circulation and cooling blood,etc.In the treatment of allergic skin diseases,The majority of prescriptions were cold drugs,and the tastes were mainly sweet and bitter,while the meridians were mainly concentrated in the liver,spleen and lung organs.The most frequently used TCM were heat-clearing drugs(such as Rehmannia glutinosa(Gaertn.)Li-bosch,Cynanchum otophyllum Schneid,Cortex Dictamni,etc.),strengthening health and dispelling pathogenic factors(such as Angelica Sinensis,Astragalus membranaceus(Fisch.)Bunge,etc.)and wind-clearing drugs(Saposhnikovia divaricata(Trucz.)Schischk,Periostracum Cicadae,Nepeta cataria L,Ligusticum chuanxiong Hort,etc.).The combination of TCM was mostly characterized by dispelling wind,cooling blood and promoting blood circulation.2.Core pescription of the new prescription can counteract the pruritus induced by caudal injection of low molecular dextran,and the anti-pruritus effect of core prescription in high dose,medium dose and low dose group is decreased,indicating That the concentration of core prescription is positively correlated with the anti-pruritus effect.3.Core prescription of the new prescription can inhibit the production of histamine(HIS),Ig E and IL-4,promote the secretion of IFN-γ,inhibit pro-inflammatory factors,and reduce the inflammatory response of mice with skin allergy induced by low molecular dextran. |
