| Objective:By observing the influence of Yuzhuozhixiao Pill on the pathological changes of liver tissue,blood biochemical indexes and the expression of key proteins NF-κBp50 and NF-κBp65 in NF-κ B signaling pathway in rats with nonalcoholic steatohepatitis(NAFLD),the mechanism of this prescription in repairing liver tissue injury was discussed.Methods: A total of 44 healthy male SPF SD rats with body weight of 180±20g,3-4 rats/cage,were fed adaptively for 1 week.Ten rats were randomly selected as blank group,and the rest were fed high fat diet(3% cholesterol +10% lard+87% basic diet)for 12 weeks.Four rats were fed high fat diet for detection.The rats fed with high fat diet were randomly divided into 3 groups(Yuturbadiaowan group,model group and positive control group),with 10 rats in each group.Except the blank group,the other 3 groups were given high fat diet.The yuturbadiaowan group and positive control group were given drug intragastric administration for 12 weeks,respectively,and killed after the last administration.The expressions of biochemical indices(AST,ALP,ALB,ALT),inflammatory cytokines(TNF-α,IL-1,IL-6),and NF-κB p50,NF-κB p65 and their phosphorylation in liver were detected.Results:1.General conditions of rats: The body weight of rats before modeling was not statistically significant(P > 0.05).There was a significant difference in the trend of body weight increase between blank group and model group in the middle of modeling week(P < 0.05),and the difference in weight gain of rats in high fatdiet group was significantly higher than that in ordinary diet group(P < 0.05).2.Liver condition in rats.2.Liver conditions of rats: compared with the blank group,liver in the model group was larger in volume,dull in luster,indicating uneven concave and convex,more granular sensation,round and dull in edge,darker in color,and yellowish in color.Liver in Yuzhuzhixiao Wan group: soft texture,fair glossiness,appearance color similar to reddish-brown,liver compared with model group: smaller volume,less granular sensation,sharp edge.3.Effect of Yuturbadixiao Pill on serum AST,ALP,ALB and ALT levels in rats with non-alcoholic fatty liver disease: compared with model group,serum AST and ALT levels in Yuturbadixiao Pill group were decreased,while ALB levels were increased(P < 0.01),and liver function was significantly recovered compared with other groups,with statistical significance.4.The effect of Yuzhuzhixiao pills on the morphology of liver tissue in rats with non-alcoholic fatty liver disease.4.The effect of Yuzhuozhixiao Pills on the morphology of liver tissue in rats with non-alcoholic fatty liver disease Results: Compared with the model group,the histomathology of Yuzhuozhixiao Pills group showed that the degree of cell swelling was significantly reduced,a small amount of fat vacuole appeared in the cytoplasm,diffuse exudation was reduced,and the number of infiltrating inflammatory cells in the junction area was reduced,which was better than the control group.5.Effect of Yuturbadixiao Pill on serum inflammatory cytokines TNF-α,IL-1 and IL-6 in rats with non-alcoholic fatty liver disease: compared with blank group,the levels of serum inflammatory cytokines TNF-α,IL-1 and IL-6in rats in model group were increased;Compared with model group,the levels of serum inflammatory cytokines TNF-α,IL-1 and IL-6 in Yuzhuzhixiao Wan group were decreased significantly(P < 0.05);The levels of serum inflammatoryfactors TNF-α,IL-1 and IL-6 in bifidobacterium group were decreased significantly(P < 0.05).6.Effect of Yuturbadixiao Pills on the expression of NF-κB p50 and NF-κB p65 protein in rats with non-alcoholic fatty liver disease: compared with blank group,the expressions of NF-κB p50 and NF-κB p65 protein in model group,Yuturbadixiao Pills group and bifidobacterium group had no significant changes(P > 0.05).After phosphorylation: compared with blank group,the expressions of P-NF-κB p50 and P-NF-κB p65 in model group were significantly increased(P < 0.05).Compared with model group,the protein expression of P-NF-κB p50 in Yuzhuzhixiao Wan group was significantly decreased(P < 0.05),which was better than that in control group.Conclusion(s):1.Yuzhuozhixiao pills can effectively improve the liver function and other biochemical indexes of NASH rats and regulate lipid metabolism,and the efficacy of TCM group is significantly better than that of model group.2.By regulating the expression of p-NF-KBp65 in liver tissue,Yuturaozhixiao pills can effectively improve liver function and other biochemical indexes,reverse the degree of steatosis in liver tissue of NASH rats,and reduce liver inflammation,which may be the mechanism of Yuturaozhixiao pills in preventing and treating NAFLD. |
