| Objectives To explore the expression level and clinical value of histone H3K36me2 in glioma cells of different grades,in order to provide a potential diagnostic marker for glioma.Methods A total of 73 patients with glioma who received surgery in Hebei general hospital were recruited.The clinical and pathological data were collected as follows:gender,age,tumor diameter,Ki-67,O6-methylguanine-DNA methyltransferase(MGMT),p53,Isocitrate dehydrogenase 1(IDH1),etc.H3K36me2 level was measured by immunohistochemical method postoperatively.The IHC results were scored and SPSS25.0 was used statistical analysis.Chi-square test was used to investigate the effects of different clinical features on H3K36me2 expression levels.Spearman correlation test was used to explore the correlation between different pathological features and H3K36me2 expression.The relationship between H3K36me2 and the prognosis of glioma was investigated by Kaplan-Meier curve and Cox regression model.Results 1 After immunohistochemical staining of glioma tissues,H3K36me2 expression was significantly different in different grades of glioma,and the difference was statistically significant(P<0.05).The expression level of H3K36me2 was the highest in grade 4 glioma and the lowest in grade 2 glioma(P<0.05).2 There was no statistical difference between high expression rate of H3K36me2 and genders(P>0.05)or tumor diameters(P>0.05).The high expression rate of H3K36me2 in older patients(>52)was significantly higher than that of younger patients(<52)(P<0.05).3 Spearman correlation test showed that IDH1 mutation resulted in decreased H3K36me2 expression(r=-0.32,P<0.05)and MGMT expression level(r=-0.35,P<0.05)were negatively correlated with H3K36me2 expression level.The expression levels of p53(r=0.38,P<0.05)and Ki-67(r=0.44,P<0.05)were positively correlated with the expression levels of H3K36me2.4 Kaplan-Meier curves suggested that higher Higher expression level of H3K36me2 in glioma tissues is correlated with lower the progression free survival(PFS)and overall survival(OS)(P<0.05).5Univariate Cox regression analysis showed that patients with older age,larger tumor diameter,higher expression of Ki-67,higher grade of glioma,higher expression level of H3K36me2,and lower expression level of MGMT,had shorter OS and PFS of patients(P<0.05).Compared with IDH1 wild-type,patients with IDH1 mutation(P<0.05)had longer OS and PFS.However,the expression level of p53 and gender had no significant influence on OS and PFS(P>0.05).Multivariate Cox regression analysis showed that tumor diameter,glioma grade,IDH1 mutation were independent prognostic factors for OS and PFS evaluation(P<0.05).High H3K36me2 expression level resulted in shorter overall survival(P<0.05),but there was no significant difference in progression-free survival(P>0.05).Conclusions 1 The expression level of H3K36me2 is different in different grades of glioma.The expression level of H3K36me2 was highest in grade 4 gliomas and lowest in grade 2 gliomas.2 The expression level of H3K36me2 was correlated with the expression level of common clinical markers such as Ki-67,p53 and MGMT,and IDH1 mutation.3The expression level of H3K36me2 is correlated with the prognosis of patients with glioma.Preliminary results suggest that H3K36me2 can be used as an independent prognostic factor to evaluate the overall survival of patients.Figure 5;Table 9;Reference 166... |
