| Objective:Sequencing of aortic dissection genes based on Poly A transcriptome sequencing technology,using bioinformatics analysis,and to identify the causative genes involved in the occurrence and development of aortic dissection(aortic dissection,AD),thus providing different new targets and ideas for continued in-depth study of aortic dissection and providing a basis for AD treatment and prognosis in the clinic.Methods:1.Whole transcriptome sequencing was performed on the aortic wall of 12 individuals with aortic dissection,as well as on normal aortic wall samples from four individuals with aortic valve disease.2.The above samples were sequenced using the K5500 and Agilent 2200 Tape Station Sequencer with PE150 mode.High quality data were obtained by standardizing the above two groups of reads using HISAT2,and the differentially expressed genes were screened by DESeq2.3.The above genes were subjected to GO and KEGG enrichment analysis and protein interaction network analysis in the GO and KEGG databases.Results:Based on the 102 gb of data obtained from transcriptome sequencing,595 upregulated and 1072 downregulated genes were identified according to the DEGs.Go enrichment analysis revealed that these differential gene were positively correlated with regulatory biological processes at the hormone level,cellular components of collagen containing extracellular matrix,and function of signaling receptor activating active molecules(P<0.05).KEGG enrichment analysis identified that these differential genes were significantly enriched mainly in PI3 K Akt signaling,calcium signaling and other pathways,after which we also constructed a protein interaction network(PPI)and screened VEGFA,IL6,FN1,IGF1,CXCL8,HIF1 A,KDR,SPP1,HGF,and CCL2 that might be pathogenic genes for AD,while the hub gene with the highest node was VEGFA and found that it was closely related to extracellular matrix.Conclusions:We performed a comprehensive analysis of aortic dissection based on Polya transcriptome sequencing and bioinformatics analysis.The findings suggest that VEGFA,IL6,FN1,IGF1,CXCL8,HIF1 A,KDR,SPP1,HGF,and CCL2 may be key differentially expressed genes for aortic dissection. |
