| Background: Bladder cancer is a common urinary system tumor with insidious onset and difficult early diagnosis.Due to its easy recurrence and metastasis,it often leads to treatment failure and death in patients with bladder cancer.About 70%-80%of new cases of bladder uroepithelial cancer are non-muscular invasive.However,50%-70% of bladder cancer still relapses after electroresection of bladder tumor combined with intravesical perfusion,and even 10%-30% of bladder cancer patients eventually develop invasion into the bladder muscular layer,leading to distant metastasis and poor treatment effect.The recurrent of bladder cancer,coupled with the risk of muscle layer progression,necessitates that its patients be monitored for life and treated repeatedly after tumor recurrence,which brings a huge financial strain on the medical system and a significant psychological strain on the patients.At present,the cause and progression of bladder cancer remain largely mysterious.This research field concentrates on the pathogenesis of bladder cancer,as well as the search for effective intervention molecular targets,in order to identify effective methods for diagnosing and treating the condition.YAP is an important downstream effector that regulates gene expression in Hippo signaling pathway.Abnormal YAP expression is involved in the occurrence,progression and metastasis of cancer.The key role of homocysteine protein cyr61 is to activate epithelial-mesenchymal transformation(EMT),thereby augmenting tumor migration and invasion,as well as stimulating tumor growth.In addition,Cyr61 is also a key downstream target gene of Hippo pathway.The YAP/TAZ/Cyr61 axis has been observed to be involved in liver fibrosis and inflammation,yet its regulation of EMT and metastasis in bladder cancer cells has not been documented.This research endeavors to investigate the regulatory effect of YAP/TAZ/Cyr61 on EMT in bladder cancer cells.Objective: To explore the mechanism of YAP/TAZ/Cyr61 axis regulating EMT in bladder cancer cells.Methods: Firstly,the expression of YAP,TAZ and Cyr61 was verified in clinical samples,then the function of YAP and Cyr61 was verified by interfering the expression of YAP and Cyr61 in human bladder cancer cells T24 and 5637,and the effect of Yap on human bladder cancer cells through Cyr61 was verified by retrospective experiments.Results: Immunohistochemical results showed that YAP,TAZ and Cyr61 were significantly up-regulated in bladder cancer tissues,and interference experiments confirmed that interference with the expression of YAP and Cyr61 could significantly inhibit the proliferation of bladder cancer cells,promote their apoptosis,and weaken their invasion ability.The retrospective experimental results also showed that interference with YAP expression would lead to increased apoptosis rate,and overexpression of Cyr61 could slightly reverse this phenomenon,but could not completely reverse it,indicating that the downstream target gene of YAP was not only Cyr61.Conclusion: The YAP/TAZ/Cyr61 axis mediates the proliferation and invasion of bladder cancer cells in bladder cancer,and then regulates the EMT of bladder cancer cells. |
