| Objective: To investigate the role of kindlin-2 in depression-like behavior induced by unpredictable chronic mild stress in mice.Method: Male C57BL/6 mice were randomly divided into control group and unpredictable chronic mild stress(UCMS)group.Forced swimming,tail suspension and sucrose preference test were used to detect depressive-like behavior of the mice.Rt-q PCR,WB,IHC and other techniques were used to observe the expression of kindlin-2 in the hippocampus of each group.IF was used to observe the cell types of kindlin-2 expression in hippocampus.Further,on the second day after the end of behavioral testing,the hippocampus was injected with overexpressed kindlin-2adeno-associated virus(AAV-Kindlin-2)or control virus(AAV-EGFP)using stereotactic technique.After 4 weeks of virus injection,the expression of kindlin-2was detected,and depressive-like behavior was detected by forced swimming,tail suspension,and sucrose preference test.Results: Compared with the control group,after 28 days of unpredictable chronic mild stress,The mice in UCMS group lost body weight,increased immobility time in forced swimming and tail suspension experiments,and decreased preference for sugar water,which suggesting depressive-like behavior appeared in mice.Kindlin-2 was mainly expressed in hippocampal neurons,and the m RNA and protein levels of kindlin-2 in hippocampus of UCMS group were significantly down-regulated compared with the control group,and the difference was significant.Overexpression of kindlin-2 in hippocampus significantly improved depressive-like behavior.Conclusion: Unpredictable chronic mild stress induced depressive-like behavior in mice,resulting in decreased hippocampal kindlin-2 m RNA and protein levels.Overexpression of kindlin-2 in hippocampus of mice ameliorated depressive-like behavior induced by UCMS.Figure 19,Table 2,References 165... |
