Stem cells from human exfoliated deciduous teeth(SHED)are mesenchymal stem cells derived from neural crest.Preclinical studies have demonstrated that mesenchymal stem cells are a potential way to treat major depressive disorder(MMD).At present,there is no report on the antidepressant effects of SHED.Objective: 1)To investigate the effect of SHED on depression-like behavior of mice;2)To evaluate the safety of SHED injection to CUMS mice.Methods: 1)Use C57BL/6J mice as the research object,establish chronic unpredictable mild stress(CUMS)mouse model,SHED was transplanted into mice by caudal vein injection.Mice were divided into four groups: normal control group,model control group,SHED group and fluoxetine group.Observe the changes of body weight of mice.Behavioral tests were performed to observe depression-like behavior changes in mice.2)To evaluate the safety of SHED injection to CUMS mice,take the body temperature of CUMS mice after SHED injection,use flow cytometry to analyze T lymphocytes in plasma and spleen in each group,and Hematoxylin-eosin(HE)staining was performed on the heart,liver,spleen,lung and kidney sections.Results: 1)After SHED injection,CUMS mice gained weight and their sugar water preference index increased,total distance of autonomous activity increased in the later period,central distance and central time increased,tail suspension immobility time decreased in the later period,recognition index increased and forced swimming immobility time decreased.The therapeutic effect of SHED lasted up to 6weeks after the end of dosing.SHED had a better effect than fluoxetine in improving autonomous activity and immobile time of forced swimming,and the difference was statistically significant(P<0.05).2)After SHED injection,the body temperature of CUMS mice fluctuated around35.5-37.5 ℃ without significant increase or decrease;There was no significant difference in the proportion of T lymphocytes in plasma and spleen(P>0.05)and HE sections of organs among all groups.Conclusions: 1)SHED treatment of CUMS mice is effective,persistent and safe;2)SHED treatment of MDD has a promising clinical application.
