Bavachin is a class of flavonoids extracted from psoralea fruit and used as a traditional Chinese herbal medicine in Asian countries.Studies have proved that Bavachin is involved in regulating the insulin sensitivity of 3T3-L1 cells,which can inhibit the occurrence and development of diseases such as inflammation and cancer,and is closely related to tumor and metabolic diseases.However,whether Bavachin is involved in the regulation of liver glucose and lipid metabolism has not been reported.Therefore,this subject mainly studies the regulation effect of Bavachin on liver glucose and lipid metabolism in vitro and in vivo.Mainly through isolating and culturing mouse liver primary cells and constructing mice models of non-alcoholic fatty liver disease.Bavachin was treated at the cell and animal levels,and q-PCR and western blot were used as the main research methods.The role of Bavachin in the regulation of insulin sensitivity and lipid metabolism in the liver at the cellular and animal levels was investigated.We found that Bavachin significantly increased the phosphorylation of insulin-induced AKT in mouse liver primary cells.In addition,Bavachin treatment significantly reduced fatty acid-induced TG synthesis in cells,showing inhibition of the expression of adipose-synthesis-related genes.In addition,in the hepatocyte steatosis model induced by high glucose and high insulin,the expression of lipid synthesis related proteins was decreased after treatment with Bavachin,and the volume and number of lipid droplets in the cells were decreased compared with the control group by oil red staining.These results were consistent in normal mouse and human liver cell lines treated with Bavachin.At the animal level,we demonstrated that Bavachin significantly improved fatty liver symptoms in mice induced by a high fat diet.Results of the IPGTT and IPITT trials confirmed that Bavachin improved glucose tolerance and insulin resistance induced by high fat diet.These results indicate that Bavachin is promising as a new drug for the treatment of metabolic diseases such as obesity,type 2diabetes and non-alcoholic fatty liver disease.
