ObjectiveTo investigate the expression level and clinical significance of serum long noncoding RNA(lncRNA)H19,CHAST in patients with coronary atherosclerotic heart disease.MethodsA total of 135 patients with coronary heart disease who were admitted to the Department of Cardiovascular Medicine,Lianyungang Second People’s Hospital,affiliated to Bengbu Medical College,from June 2021 to May 2022 were selected,including 76 cases of acute coronary syndrome(ACS)and 59 cases of chronic coronary syndrome(CCS).Meanwhile there were 62 patients in the control group who were hospitalized and excluded coronary heart disease by coronary angiography.Real-time fluorescent quantitative PCR(RT-PCR)was used to decet the relative expression level of lncRNA H19,lncRNA CHAST in the circulating serum of the subjects.Receiver operating characteristic(ROC)curve was used to analyze the clinical diagnostic value of circulating serum lncRNA H19 and lncRNA CHAST expression level in the diagnosis of coronary heart disease.Pearson correlation coefficient was used to analyze the correlation between serum lncRNA H19,lncRNA CHAST level and hs-CRP and TNF-α,IL-6,LDL,HDL and Gensini scores.Multivariate logistic regression was used to analyze the risk factors of coronary heart disease.ResultsThere was no significant difference in age,BMI,history of diabetes,drinking history,TG,TC and other indicators between ACS group,CCS group and control group(P>0.05).Compared with the control group,sex,smoking,hypertension,LDL,HDL,hs-CRP,TNF-α,IL-6 and other indicators in ACS group and CCS group were significant differences(P<0.05).Compared with CCS group,the expression level of LDL,hs-CRP,TNF-α and IL-6 in ACS group was significantly higher(P<0.05),while the expression level of HDL was lower(P<0.05).The relative expression level of serum lncRNA H19 and lncRNA CHAST in the coronary heart disease group was higher than that in the healthy control group,and the relative expression level of lncRNA H19 and lncRNA CHAST in the ACS group was higher than that in the CCS group,the difference was statistically significant(P<0.05).The expression levels of serum lncRNA H19 and lncRNA CHAST were positively correlated with the hs-CRP,IL-6,TNF-α,LDL and Gensini scores in patients with coronary heart disease,and negative correlated with HDL expression level.(P<0.05).The ROC curve analysis showed that lncRNA H19 and lncRNA CHAST had good diagnostic value as the diagnostic index of coronary heart disease,AUC was 0.9025 and 0.7749 respectively,and the combined diagnosis of the two was more valuable,AUC was 0.9299.Multivariate logistic regression analysis showed that hypertension,smoking,hs-CRP,LDL,lncRNA H19,lncRNA CHAST were independent risk factors for coronary heart disease(P<0.05).Conclusions(1)The expression levels of LncRNA H19 and lncRNA CHAST in the serum of patients with coronary heart disease were significantly increased.(2)The serum expression levels of LncRNA H19 and lncRNA CHAST in patients with coronary heart disease are related to inflammatory markers and lipids.(3)The expression levels of serum lncRNA H19 and lncRNA CHAST in patients with coronary heart disease can reflect the severity of coronary artery disease to a certain extent.
