Effect Of Adipose-derived Mesenchymal Stem Cells On Hematopoietic Support And Graft-versus-host Response Based On NOD/SCID Mice

Objective:Adipose-derived mesenchymal stem cells have unique biological properties and a wide range of sources,while whether they support hematopoiesis in vivo has not been fully determined.This study investigates whether they support hematopoiesis in vivo and their effects on graft-versus-host responses based on the NOD/SCID mouse hematopoietic stem cell transplantation model.Materials and methods:BMNCs and ADSCs cells of male C57BL/6 mice were obtained in isolation and culture,and female NOD/SCID mice were used as recipients,which were irradiated with sublethal doses of X-rays(3Gy)and divided into 2 groups:(1)The control group was infused with 1×10~6BMNC cells via tail vein.(2)1×10~6BMNC cells and 1×10~6ADSC cells were infused into the tail vein of the experimental group.Both groups of mice were monitored for body weight and blood count after transplantation,and the expression of the specific gene SRY on the Y chromosome of bone marrow cells was detected by quantitative PCR,and pathological histological analysis of liver,lung,spleen,intestine and bone was performed.Results:(1)There was no significant difference in body weight change between the 2 groups after transplantation(P>0.05);(2)There was no significant difference in white blood cell count between the 2 groups after transplantation,0.258±0.760(10~9/L)in the control group and 0.218±0.046(10~9/L)in the experimental group in the first week,P>0.05;0.280±0.099(10~9/L)in the control group and 0.298±0.107(10~9/L)in the experimental group in the second week,P>0.05;1.518±1.032(g/L)in the control group and 1.162±0.721(10~9/L)in the experimental group in the fourth week,P>0.05.(3)In terms of platelet recovery,it was 316.0±17.349(10~9/L)in the control group and 298.666±19.296(10~9/L)in the experimental group in the first week,P>0.05;it was308.0±44.508(10~9/L)in the control group and 321.0±54.442(10~9/L)in the experimental group in the second week,P>0.05.In the fourth week,it was 624.67±67.988(10~9/L)in the control group and 793±32.234(10~9/L)in the experimental group,P=0.018(P<0.05).week four was 624.67±67.988(10~9/L)in the control group and 793±32.234(10~9/L)in the experimental group,P=0.018(P<0.05),the platelet count was higher in the experimental group than in the control group.(4)In terms of hemoglobin recovery,it was 71.333±4.509(g/L)in the control group and 74.333±2.517(g/L)in the experimental group in the first week,which was greater than the control group,but P>0.05;it was 64.0±3.606(g/L)in the control group and 58.666±2.082(g/L)in the experimental group in the second week,P>0.05;it was 88.333±3.786(g/L)in the control group and 84.0±4.0(g/L)in the experimental group in the fourth week,P>0.05,with no significant difference.(5)SRY gene expression was detected in the Y chromosome in both control and experimental groups,1.268±0.149(%)in the experimental group and 0.914±0.111(%)in the control group,which was higher in the experimental group than in the control group,P=0.014(P<0.05),indicating that ADSCs can accelerate chimeric implantation of donor stem cells.(6)In the histopathological analysis,the experimental group had a relatively mild degree of tissue damage and relatively little inflammatory cell infiltration,with a histopathological score of P<0.05 between the 2 groups.Conclusions:(1)ADSCs have the effect of promoting platelet recovery;(2)ADSCs can promote donor stem cell implantation;(3)ADSCs attenuate graft-versus-host reaction after transplantation.
