| Objective:To investigate the relationship between RELN and prognosis in patients with colorectal cancer.Methods:We first screened out the differentially expressed gene RELN in the TCGA database,and verified the expression of RELN and its relationship with the prognosis of colorectal cancer patients in the TCGA database.Cox regression method was used for multivariate analysis,and then Nomogram was constructed on this basis for internal verification.The correlation between RELN and immune cells,immune-related expressed genes and microsatellite instability molecules were analyzed in the TIMER database.Then,the correlation between RELN and immune checkpoint related molecules was explored in GEPIA database.Finally,the possible correlation pathways were analyzed by GSEA.At the same time,30 patients with colorectal cancer admitted to the Department of General Surgery of the Northern Theater Command General Hospital of the People’s Liberation Army from March 2022 to June 2022 were selected,and all patients’ postoperative resection of colorectal cancer tissues and adjacent tissues were retained,and the expression level of RELN in colorectal cancer was verified by PCR.Results:According to database screening,the differentially expressed molecule is RELN.The expression of RELN in paracancer tissues is higher than that in cancer tissues.Colorectal cancer patients with high RELN expression have better prognosis.Then patients with low RELN expression COX regression analysis showed that RELN expression and M stage,lymphatic invasion were independent risk factors for the progression of colorectal cancer.In the internal verification,the C-index of the nomogram was 0.8493(95%CI 0.7735-0.9251),indicating that the prediction model had good accuracy.The gene enrichment method found that RELN affects the biological events of colorectal cancer by affecting calcium signaling pathway,DNA methylation,complement cascade reaction,etc.The TIMER database showed that RELN expression was significantly correlated with immune infiltration,and may play an important role in immune escape of tumor cells in the CRC microenvironment.PCR results showed that the expression level of RELN in colorectal cancer tissue samples was lower than that in paired paracancer tissues.Conclusions :Elevated RELN can significantly prolong OS in colorectal cancer patients,and can be used as a novel predictive biomarker for colorectal cancer.It may also be associated with immune infiltration of colorectal cancer,and may play an important role in the occurrence of immune escape of tumor cells in the colorectal cancer microenvironment.Further PCR to elucidate the value of RELN gene in colorectal cancer are critical. |
