Objective: To construct a radioimmune cell line for lung adenocarcinoma and detect the difference in glycolysis levels between the radioresistant cell line and the parent strain of lung adenocarcinoma.Using glycolysis inhibitor 2-DG to evaluate radiosensitization effect on the radioresistant cell line of lung adenocarcinoma.Methods: By gradient dose irradiation,radioresistant cell lines of lung adenocarcinoma were constructedand.Comparing the radiotherapy sensitivity differences between radioresistant cell lines and parent strains by clonal formation experiments,and the expression differences of stem proteins were detected.Successfully constructed cell lines were selected and the expression differences of key enzymes of glycolysis at the protein and RNA levels were compared by Western Blot experiment and Real-time PCR experiment.Lactate and glucose content were measured to compare the differences in metabolic levels.2-DG was used to treat the radioresistant cell line of lung adenocarcinoma,CCK8 experiment observed cell proliferation,cloning formation experiment compared the change of radiotherapy sensitivity,and judged whether 2-DG had the effect of reversing radioresistance.Results: 1.Cloning formation experiments and stem protein level detection verified the successful construction of lung adenocarcinoma radioimmune cell lines.2.Compared with the parent strain,glycolysis-related enzymes HK2,PKM2 and GLUT1 increased at the RNA and protein levels,lactic acid production increased,and glucose consumption was not significantly different.3.The use of glycolysis inhibitor 2-DG can increase the radioresistance to the death of lung adenocarcinoma cells after irradiation and improve radiosensitivity.Conclusions: 1.The level of glycolysis of radioresistant cell lines of lung adenocarcinoma is increased,and the radiosensitivity is reduced.2.Glycolysis inhibitor2-DG can improve radiosensitivity of radioresistant lung adenocarcinoma cell lines and has a reversible effect on radioresistance.
