Objective: Based on the data of the TCGA online database,the target gene that promotes the progression of non-small cell lung cancer(NSCLC)was found,and the relationship between the expression of this gene and the prognosis of NSCLC was explored,so as to explore the therapeutic targets related to target genes.Methods: The expression data of 1102 cases of VAV2 m RNA in NSCLC and the clinical relevant data of patients were downloaded from the TCGA database(https://portal.gdc.cancer.gov/),and the R4.2.2 software was used to analyze the expression differences of VAV2 m RNA in NSCLC tumor tissues and paracancerous tissues through t-test,and the Log-rank test for VAV2 Survival analysis was performed in NSCLC patients with different m RNA expression,the effect of VAV2 m RNA and clinicopathological features on the prognosis of NSCLC patients was analyzed by univariate and multivariate COX,the effect of VAV2 on NSCLC tumor microenvironment and tumor mutation burden was analyzed by nonparametric tests,and the relationship between the expression of VAV2 in NSCLC tissues and the degree of invasion of immune-related cells was analyzed by using the Timer database.The TCIA database was used to analyze the effect of VAV2 on NSCLC immunotherapy.Results: 1.Upregulation of VAV2 m RNA expression in NSCLC tumor tissues.2.The overall survival and progression-free survival of NSCLC patients were positively correlated with VAV2 m RNA expression levels.3.VAV2 can be independent of other factors as an independent prognostic factor for NSCLC.4.The content of stromal cells and immune cells in the tumor microenvironment of NSCLC patients with high VAV2 expression was higher than that of patients with low expression.5.In the tumor microenvironment of NSCLC patients,the level of regulatory T cell infiltration(r=0.22,P<0.001)and the resting memory CD4+ T cell infiltration level(r=0.086,P<0.05)were positively correlated with VAV2 m RNA expression level,macrophage M2 type infiltration level(r=-0.089,P<0.05),resting NK cell infiltration level(r=-0.099,P<0.05),The resting dendritic cell infiltration level(r=-0.097,P<0.05)and eosinophil infiltration level(r=-0.081,P<0.05)were negatively correlated with VAV2 m RNA expression level.6.We found CLTA4,CD44,CD40 LG,TNFRSF25,ADORA2 A,TNFRSF4,PDCD1,TNFSF15,TNFRSF14,TNFRSF9,NRP1,VTCN1,CD276,TNFSF18,TNFSF14,LAIR1,ICOS,TIGIT,CD160,CD28,TMIGD2,TNFRSF8,CD70,ICOSLG,Immune checkpoint genes such as LAG3 have a positive regulatory relationship with VAV2 7.NSCLC patients with high VAV2 m RNA expression will have better efficacy when receiving immunotherapy.Conclusions: 1.VAV2 m RNA is highly expressed in NSCLC.2.High expression of VAV2 m RNA is associated with poor prognosis of NSCLC.3.VAV2 can affect the tumor microenvironment of NSCLC patients by affecting their immune cell infiltration.4.This study provides a new idea for the application of immunosuppressants in NSCLC.
