Objective:Based on DSP and n Counter platform found primary tumors and Cervical lymph nodes metastases in patients with nasopharyngeal carcinomaand lung metastases tumor immune microenvironment difference between;Using bioinformatics screen interact with the immune microenvironment and genetic variations affect the occurrence and development of nasopharyngeal carcinoma.Methods:Choose between January 2020 and December 2022 in affiliated hospital of Hainan medical college(Hainan General Hospital),department of nasopharyngeal carcinoma of the primary lesion and neck lymph node metastasis lesions paraffin samples are divided into primary focal nasopharyngeal carcinoma and cervical lymph node metastases group;Choose nasopharyngeal carcinoma patients of primary lesion and lung metastatic lesions of paraffin samples are divided into primary nasopharyngeal carcinoma and lung metastases group oven.Using Nano String’s Geo Mx Digital Spatial Profiling(DSP)platform and n Counter platform for data analysis,differential genes related to the tumor immune microenvironment were identified using a threshold of False Discovery Rate(FDR)value<0.05 and an absolute valueof log2(Fold Change)>1.Choose 20 nasopharyngeal carcinoma with metastatic lesions from the primary lesion of matching cases and 10 cases of normal nasopharyngeal tissue contrast nasopharyngeal carcinoma by immunohistochemical method of primary tumors and cervical lymph node metastases group differences in gene validation;Choose 6 cases of pulmonary metastasis of nasopharyngeal carcinoma,nasopharyngeal carcinoma,22 cases from the primary lesion and 10 normal nasopharyngeal tissue contrast cases nasopharyngeal carcinoma by immunohistochemical method of primary tumors and lung metastases group differences in gene validation.Results:Application of DSP and n Counter platform analysis of primary nasopharyngeal carcinoma(NPC)focal lesions and cervical lymph node metastases group,a total of 9is analyzed with the statistically significant(P<0.05),the difference of gene(raised gene 8,down gene 1);Further using immunohistochemical methods validation difference value of the biggest MMP1 and PDK1 in primary nasopharyngeal carcinoma(NPC)expression of focal lesions and cervical lymph node metastases group,after statistics analysis,neck lymph node metastasis lesions than high expression from the primary lesion and statistically significant(P<0.05).For nasopharyngeal carcinoma primary tumors and lung metastatic lesions group a total of 26 is analyzed with the statistically significant(P<0.05).The difference of gene(raised gene23,down gene3);further using immunohistochemical methods validation difference value of the biggest FN1 in nasopharyngeal carcinoma primary tumors and the expression of pulmonary metastatic lesions group,after statistics analysis,the primary lesions than lung metastases high expression and statistically significant(P<0.05).Conclusion:Compared with the primary focal nasopharyngeal carcinoma,cervical lymph node metastases tumor immune microenvironment difference,MMP1 and PDK1 in cervical lymph node metastases,high expression may be associated with cervical lymph node metastases nasopharyngeal carcinoma.Compared with the primary focal nasopharyngeal carcinoma,lung metastases tumor immune microenvironment difference,FN1 in primary focal high expression,may be associated with lung metastasis occurred.
