| Objective: To explore the correlation between the expression level of Hsa_circ_0039351 and the clinicopathological data of colorectal cancer patients,and further clarify its biological function in the occurrence and development of colorectal cancer.Methods: The 53 cases of surgical resection of colorectal cancer and paracancerous tissues from the Department of Gastrointestinal Surgery of our hospital were collected as research objects.High-throughput sequencing was used to construct three pairs of circ RNA differential expression profiles of colorectal cancer tissue and adjacent normal intestinal epithelial tissue.The three circ RNAs with the most obvious expression differences were selected,and their expression levels were verified by RT-q PCR in 50 pairs of CRC tissues,and the Hsa_circ_0039351 was determined as the target gene of this study.The t-test was used to analyze the correlation between Hsa_circ_0039351expression level and the clinicopathological data of rectal cancer.RNase R experiments were used to verify the stability of the Hsa_circ_0039351.Verify Hsa_circ_0039351expression levels in colorectal cancer cell lines and construct knockdown and overexpression cell lines.The effects of overexpression and knockdown on the proliferation of rectal cancer cells were detected by CCK-8 experiment and Ed U experiment Hsa_circ_0039351,the effects of overexpression and knockdown Hsa_circ_0039351 on the migration of colorectal cancer cells were detected by transwell experiment and cell scratch experiment,and the effect of overexpression and knockdown Hsa_circ_0039351 on apoptosis of rectal cancer cells was detected by flow cytometry.Graph Pad Prim 8.0 software was used for statistical analysis and mapping.Statistical analysis of experimental data was performed using t-test.The data are expressed as mean ±SD and P<0.05 is considered statistically significant.*p<0.05,**p<0.01,***p<0.001。Results: The results of high-throughput sequencing showed that a total of 171 circ RNAs were highly expressed in colorectal cancer and adjacent tissues,among which the expression of Hsa_circ_0001681,Hsa_circ_0039351 and Hsa_circ_0063865 was the most obvious.Further verification in 50 pairs of cancers and adjacent tissues showed that the expression level of Hsa_circ_0039351 in colorectal cancer tissues was significantly higher than that of normal tissues(p<0.001).There was a significant correlation between the expression level of Hsa_circ_0039351 in colorectal cancer organizers and tumor size,tumor invasion,lymph node metastasis and vasculature invasion in colorectal cancer patients(p<0.05).Functional experiments showed that knocking down the expression of Hsa_circ_0039351 in colorectal cancer cells could significantly reduce the proliferation and migration ability of colorectal cancer cells,and their apoptosis rate was significantly increased(p<0.05).After constructing Hsa_circ_0039351 overexpressed cell lines,it was found that the proliferation and migration ability of colorectal cancer cells were significantly improved compared with those of normal cells,and the apoptosis rate decreased(p<0.05).Conclusion: Hsa_circ_0039351 is highly expressed in colorectal cancer,and its expression level is correlated with the clinicopathological data of colorectal cancer patients.At the same time,as a cancer-promoting gene,it promotes the proliferation and migration ability of colorectal cancer cells.Hsa_circ_0039351 is expected to become a new target or biomarker for molecularly targeted therapies for the diagnosis and treatment of colorectal cancer. |
