Objective: Through searching relevant literatures of TCM phlegm-based treatment of COPD in CNKI,Wanfang and Weipu databases,sorting out TCM prescriptions,summarizing the core drug composition of TCM phlegm-based treatment of COPD,and further exploring the core target and mechanism of action of core drug composition in the treatment of COPD through network pharmacology.Methods:1.Data mining: From January 2004 to December 2021,literature related to TCM phlegm-based treatment of COPD was retrieved from CNKI,Wanfang and Weipu databases.TCM prescriptions were sorted out according to the standard of inclusion and expulsion,the names of TCM were standardized,and the sorted data were input into Excel spreadsheet to establish the literature research database of TCM phlegm-based treatment of COPD,which was imported into the cloud platform of ancient and modern medical records.After further standardized processing,the data mining method was used to analyze the traditional Chinese medicine prescription and screen out the core drug composition.2.Network pharmacology: Search and screen the active ingredients of core drugs through the TCMSP database,standardize the active ingredients in the Uniprot protein database,and obtain the current target of core drugs;Targets related to COPD were obtained by Gene Cards database,OMIM database and TTD database.Cytoscape 3.8.2 software was used to draw drug-disease target Venn diagram to obtain drug-disease common targets,construct drug active ingredient-disease target network,and obtain drug core active ingredients and key targets through topological analysis.GO and KEGG analysis of the common targets of core drugs and COPD were performed using Metascape database.The binding activity between core drug ingredients and core target was verified by molecular docking.Results:1.According to the exclusion criteria,587 literatures and 602 TCM prescriptions were selected and included.The data mining function of ancient and modern medical records platform was used to analyze the prescriptions of traditional Chinese medicine.The 602 prescriptions of Traditional Chinese Medicine involved 237 kinds of traditional Chinese medicine,and the total use frequency was 6805 times.The top 10 Chinese medicines according to the use frequency were Gancao,Banxia,Xingren,Huangqin,Fuling,Chenpi,Gualou,Sangbaipi,Mahuang,Jiegeng.The four properties are mainly "warm,cold and flat",the five flavors are mainly pungent,sweet and bitter,and the meridians are mainly lung,spleen and stomach.Cluster analysis determined four core drug combinations: Gancao,Banxia,Fuling,Chenpi;Xingren,Mahuang,Huangqin,Sangbaipi,Gualou,Zhebeimu;Zisuzi,Laifuzi,Baijiezi;Dilong,Taoren,Danshen,Yuxingcao,Tinglizi,Ziwan,Zisuzi,Houpo.Through the correlation analysis of Traditional Chinese Medicine and complex network analysis,the drug combinations with strong correlation were "Chenpi,Banxia","Fuling,Banxia","Jiegeng,Gancao","Fuling,Gancao","Mahuang,Gancao" and so on.Finally,the core drugs were selected as "Gancao,Banxia,Xingren,Huangqin,Fuling,Chenpi,Gualou".2.175 core drug active ingredients and 260 targets were screened by TCMSP database,1378 targets related to COPD were screened by Gene Cards database,OMIM database and TTD database,and 104 common targets were obtained by the intersection of drug and disease targets.The topological structure analysis of the constructed network showed that the core pharmaceutical chemical components were quercetin,stigmasterol,narengin,β-sitosterol,baicalin,kaempferol,and the key targets were STAT3,TNF,AKT1,TP53,RELA,MAPK8,CRP,MAPK1,IL6,MAPK14.Through enrichment analysis,it was found that the main signaling pathways in the treatment of COPD with core drugs include AGE-RAGE signaling pathway,PI3K-Akt signaling pathway and NF-κB signaling pathway.Conclusion:1.The core prescriptions selected in this study are: "Gancao,Banxia,Xingren,Huangqin,Fuling,Chenpi,Gualou".In the phlegm-based treatment of COPD,the disease location is mainly in the lung and spleen,and the treatment focuses on clearing heat and eliminating phlegm,regulating qi and relieving cough.2.The core prescription was analyzed through the method of network pharmacology,and it was concluded that the core prescription may participate in the treatment of COPD by inhibiting inflammation,anti-oxidation,regulating apoptosis,suppressing immune response and other mechanisms,providing certain ideas for the selection of traditional Chinese medicine for the treatment of COPD from phlegm.
