| Purpose: To observe the expression of rno_circRNA_0003340 in the liver of type 2 diabetes mellitus(T2DM)rats,analyze the relationship between rno_circRNA_0003340 and common biochemical indexes related to glucose and lipid,and explore the relationship between rno_circRNA_0003340 and T2 DM.Methods: 1.In this study,a rat model of T2 DM was established by high-fat diet combined with low-dose STZ.Sixty 5-week-old male SD rats were divided into normal control group(NC group,20 rats)and T2 DM model group(T2DM,40 rats)according to the ratio of 1:2.The rats’ weight,water consumption and urine volume were measured at 4 weeks and 8 weeks after modeling.2.At 4 weeks after modeling,all rats underwent intraperitoneal glucose tolerance test(IPGTT)and insulin tolerance test(ITT).3.The serum FPG,TG and TC of the rats were detected by an automatic biochemical analyzer,LDL-C,HDL-C,ALT,AST levels,at 4 weeks and 8 weeks after modeling.4.The levels of serum insulin(FINS)and Hb A1 c were measured by ELISA at 4 and 8 weeks after modeling.5.At 8 weeks after modeling,HE staining was performed on rat liver to observe its pathological morphology.6.At 4 and 8 weeks after modeling,q RT-PCR method was used to detect liver The expression of rno__circRNA__0003340 in tissues.7.Bioinformatics analysis of the biological functions and enriched biological pathways of rno_circRNA_0003340.Results: 1.At the 4th and 8th week,compared with the NC group,the drinking water and urine volume of T2 DM rats increased significantly,and their weight decreased significantly(P < 0.01).At the 4th week,the IPGTT-AUC and ITT-AUC in T2 DM group were higher than those in NC group(P < 0.01).2.At 4th and8 th week,FPG,Hb A1 c,TG,TC,LDL-C,ALT and AST in T2 DM group were higher than those in NC group(P < 0.01),and HDL-C in T2 DM group was lower than that in NC group at 8th week(P < 0.05).3.HE staining of liver showed that compared with NC group,the structure of hepatic lobules in T2 DM group was fuzzy,the liver turbulence is not orderly arranged,and fat droplets can be seen in the upper right of the portal area,at the 8th week.4.At 4 weeks,there was no statistical difference in the expression of rno_circRNA_0003340 between NC group and T2 DM group(P > 0.05).At the 8th week,the expression of rno_circRNA_0003340 in T2 DM group was lower than that in NC group(P < 0.05).5.Correlation analysis showed that the expression level of rno_circRNA_0003340 was negatively correlated with FPG,TG,TC and Ty G index(P < 0.05).6.Bioinformatics analysis shows that rno_circRNA_0003340 is involved in PI3K-Akt,endocrine resistance,Fox O and other signal pathways.Conclusions: 1.rno_circ_0003340 is differentially expressed in the liver tissue of rats with different course of type 2 diabetes,which may be involved in the occurrence and development of T2 DM.2.The expression level of rno_circ_0003340 in liver tissue is closely related to blood lipid,blood sugar and Insulin resistance in type 2 diabetic rats. |
