【Objective】The purpose of this study was to determine the expression of ATAD2 gene in endometrial cancer tissue specimens and paracancer tissue specimens by comparing the tissue specimens of endometrial cancer and paracancer tissue specimens,and to clarify the role of ATAD2 in the prognosis of endometrial cancer and some of its mechanisms by combining with the biogenic analysis of public databases.【Method】In this study,RNA expression data of EC were first downloaded using the American Cancer Genome Atlas(TCGA).Different mRNA,miRNA and lncRNA were screened,and Kaplan-Meier survival analysis was used to analyze the relationship between ATAD2 expression and prognosis of EC patients.Secondly,the mRNA and protein levels of ATAD2 in EC tissues were detected by RT-q PCR and immunohistochemistry,so as to explore the expression of ATAD2 in EC and paracancer tissues.The predictive value of ATAD2 as EC diagnostic factor was further analyzed by ROC method.Then GSEA software was used to analyze the functional pathway of ATAD2 gene involved in EC.The biological function,effect on immune microenvironment and drug sensitivity of ATAD2 were also analyzed.Finally,we constructed a ceRNA network of lncRNA-miRNA-ATAD2 to explore the molecular mechanism regulating ATAD2 expression.【Result】According to the analysis of the endometrial cancer data set in the TCGA database,the expression of ATAD2 in EC tissues was higher than that in paracancer tissues,and the overall survival of EC patients with high expression of ATAD2 was lower.RT-q PCR showed that ATAD2 mRNA expression increased in EC tissue,and immunohistochemical results showed that ATAD2 positive expression rate was higher in EC tissue.ROC analysis was used to further verify that ATAD2 had high predictive ability for EC diagnosis.At the same time,we found that ATAD2 expression was correlated with immune cells in the EC microenvironment.By GSEA analysis,we found that the high expression of ATAD2 mRNA was related to the changes of various signaling pathways.Drug sensitivity analysis showed that the expression level of ATAD2 was different,and the action of first-line chemotherapy drugs such as cisplatin and paclitaxel was also different.The maladjustment of ATAD2 in EC may be controlled by ceRNA network.【Conclusion】This study indicated that ATAD2 was highly expressed in patients with endometrial cancer and was associated with poor prognosis,which could be used as a prognostic indicator of EC.Biological analysis showed that the expression of ATAD2 was closely related to immune cells in the endometrial carcinoma microenvironment.At the same time,the gene expression is related to a variety of signaling pathways.Based on the conclusions of this study,it is concluded that ATAD2 can be used as an immunotherapeutic target to improve the survival and prognosis of patients with EC,and the ceRNA regulatory network of ATAD2 has potential diagnostic and therapeutic value for EC.Drug sensitivity test suggested that first-line chemotherapy drugs such as cisplatin and paclitaxel had better therapeutic effect in patients with endometrial cancer with high ATAD2 expression.
