Objective: Although the combination of surgery and neoadjuvant chemotherapy has made significant contributions,the treatment of patients with lung metastasis and local infiltration of osteosarcoma is still challenging.The pathogenesis of this cancer is still unclear.Therefore,finding new biomarkers for the diagnosis,prognosis and treatment of osteosarcoma is still an important but unmet clinical need.Subjects and Methods: Use the database to analyze the effect of RUNX2 on the KM survival curve of osteosarcoma patients and target the miRNAs related to RUNX2;Using Lipo2000,miRNAs were transiently overexpressed in osteosarcoma cell lines U2 OS and MG-63;The lentivirus transfection was used to construct a stable cell line overexpressing RUNX2;CCK-8 detection and scratch test were used to determine cell proliferation and migration respectively;Western blot analysis was performed to verify the effect of miR-758 on osteosarcoma cell line-related protein.Result: The miR-758 in the three potential target miRNAs predicted by the database could down-regulate the expression of RUNX2 in osteosarcoma cells;Mi R-758 inhibits the proliferation and migration of osteosarcoma cells U2 OS and MG-63;We have successfully constructed osteosarcoma cell lines U2 OS and MG-63 that overexpress RUNX2,and miR-758 down-regulates RUNX2 of these two cell lines;Mi R-75 downregulates RUNX2 expression by targeting the MDM2-P53 pathway.Conclusions :Our work indicates that high levels of RUNX2 can lead to poor prognosis in patients with osteosarcoma.Secondly,miR-758,as a tumor suppressor factor,can inhibit the proliferation and migration ability of osteosarcoma cells U2 OS and MG-63.This effect may be achieved by targeting the MDM2-P53 pathway and downregulating the expression of RUNX2.Finally,we believe that miR-758 may become a potential new prognostic marker and therapeutic target for osteosarcoma.
