| Objective: To explore the intervention effect of cervical vein freckle removal granules on the pathological changes of carotid atherosclerosis and related disease factors at different concentrations by establishing an Apo E-/-mouse carotid atherosclerosis model,and to provide a scientific basis for the clinical promotion of traditional Chinese medicine formulas.Methods: After rearing 55 6-week-old Apo E-/-mice with normal diet for one week,they were randomly divided into ordinary food group(5 mice,group A)and model group(50mice),and 3 mice in the model group were randomly selected with HE staining to observe the carotid artery wall of the model group after 12 weeks of feeding with high-fat diet,and the remaining mice in the model group were randomly divided into high-dose group of traditional Chinese medicine(group B),medium-dose group of traditional Chinese medicine(group C),low-dose group of traditional Chinese medicine(group D),western medicine group(group E).Each group were 8 mice.Mice in the group A and group Fwere given saline gavage,while mice in the B,C,D,E group were given daily gavage with corresponding drugs(high,medium and low doses of Jingmaiquban Granules,simvastatin)for a total of 4 weeks.After the experiment,the mice were sacrificed,the changes of body weight and length of mice were observed and calculated,and serum TC,TG,HDL-C,LDL-C were determined by enzymatic method.Determination of TNF-α,CRP with ELISA;glucose oxidase method to determine blood glucose;HE staining and oil red O staining were used to observe the pathological morphological changes of carotid arteries.Results: The differences in body weight between the treatment groups were statistically significant(P<0.05),and with the increase in drug dose,the body weight of the mice gradually decreased.Compared with group A mice,the serum of model group mice TNF-α,The expression of CRP,TC,TG,and LDL-C increased(P<0.001),while the expression of HDL-C decreased(P<0.001).Compared with the model group,the serum of mice in each treatment group TNF-α,CRP,TC,TG,and LDL-C were decreased.Comparing the differences,the results are: A<E<D<B<C<F.The difference between HDL-C can be known,and the results are: A>E>D>B>C>F.HE staining and oil red O staining showed that the lipid accumulation and plaque area of the carotid artery in the model group mice were significantly increased,and after treatment with Jingmaiquban Granule,each treatment group had different degrees of relief.Conclusion: Jumaiquban Granule has obvious effect of alleviating atherosclerosis,and its mechanism may be to reduce serum TC,TG,HDL-C,LDL-C levels,play a role in lowering blood lipids,and down regulate CRP,TNF-α.It can protect blood vessels from inflammatory damage to eliminate carotid plaque,reduce the speed of fat metabolism,and achieve the therapeutic effect of alleviating atherosclerosis. |
