| Background:Colorectal cancer is one of the most common malignant tumors in the world.With the change of living environment and habits,the incidence and mortality of colorectal cancer are increasing gradually.Through the study of many different factors involved in the development of colorectal cancer,it is believed that early screening and active treatment can improve the survival rate of patients with colorectal cancer.However,the number and specificity of the markers used for early screening diagnosis and prognosis are low.Therefore,it has become the focus of clinical scientific research to detect the potential early screening markers and study the correlation between the markers and clinicopathological parameters.The aim of this study was to analyze the expression of NUDT4 in colon cancer cells and colon cancer tissues,and to discuss the correlation between NUDT4 and PNI and other clinicopathological features,to investigate the clinical significance of NUDT4 and PNI in the prognosis of patients with colon cancer.Methods:1.The expression levels of NUDT4 in colon cancer cells,normal colon tissues and colon cancer tissues were detected by fluorescence quantitative PCR and western blotting respectively;The expression of NUDT4 in 90 cases of colon carcinoma and normal tissues adjacent to carcinoma was detected by immunohistochemical staining.2.According to the expression results of NUDT4 that detected by immunohistochemical staining,through observing the intensity of staining and the proportion of positive cells to judge comprehensively,they were divided into high expression group and low expression group,then collect clinical data and blood parameters of colon cancer patients,to investigate the effect of NUDT4 on the survival rate of colon cancer and the correlation between NUDT4 and clinicopathologic factors.The cut-off value of Prognosis Nutrition Index was determined by Receiver Operating Characteristic curve,and the influence of PNI on the survival rate of colon cancer patients and the correlation between PNI and clinicopathological characteristics were analyzed statistically,then the correlation between NUDT4 expression and PNI was analyzed.Results:1.Compared with NCM460 cells,the expression level of NUDT4 in colon cancer cells was significantly increased(P < 0.01),and the expression level of NUDT4 in colon cancer tissues was also significantly higher than that in corresponding adjacent normal colon tissues(P < 0.01).There were 62 cases with high expression of NUDT4 and 28 cases with low expression of NUDT4 in 90 cases of colon cancer,35 cases with high expression of NUDT4 and 55 cases with low expression of NUDT4 in adjacent normal tissues.2.Survival analysis showed that the overall survival rate of the high NUDT4 expression group was significantly lower than that of the low NUDT4 expression group(P = 0.003)The difference of NUDT4 expression was significant in the depth of infiltration(T stage),lymph node metastasis(N stage)and PNI(P <0.05),there was no significant difference in age,sex,TNM stage,differentiation and tumor location(P > 0.05).Univariate analysis showed that TNM stage and NUDT4 expression were risk factors for the prognosis of patients with operable colorectal cancer(P <0.05),Multivariate analysis showed that TNM stage and NUDT4 expression were independent risk factors for the prognosis of patients with operable colorectal cancer(P < 0.05).PNI cut-off value was 44.275,PNI≥44.275 was classified as high score group,PNI <44.275 was classified as low score group.Survival analysis showed that the overall survival rate of high score group was significantly higher than that of low score group(P = 0.007),PNI was significantly different in age,depth of invasion(T stage)and tumor location(P <0.05),there was no significant difference in sex,lymph node metastasis(N stage),TNM stage and differentiation(P > 0.05).Conclusions:1.The expression of NUDT4 in colon cancer was significantly up-regulated.2.The overall survival rate of low NUDT4 expression group was higher than that of high NUDT4 expression group.NUDT4 may be an independent risk factor for the prognosis of colon cancer patients.The expression level of NUDT4 was correlated with clinicopathological factors(tumor infiltration depth,lymph node metastasis,PNI value).And PNI ≥44.275 had a positive effect on the prognosis of colon cancer. |
