| Objective: The original Tibetan Mu tong base herbs were sorted into varieties,and the active parts and index components were screened based on their traditional efficacy.On this basis,the existing quality standards were improved to provide a reference for ensuring the safe and effective clinical use of Tibetan Mu tong.Methods:(1)To determine the basic medicinal materials of Tibetan mu tong based on the literature,resource investigation and chemical composition analysis.(2)Based on the traditional efficacy of Tibetan mu tong,the rat model of acute gastric mucosa injury was used to explore the active fraction and mechanism of Tibetan mu tong.(3)To study the chemical composition of the active fraction of Clematis brevicaudata DC.The compounds were separated and purified by Silica gel,Sephadex LH-20 column chromatography,preparative HPLC and semiprepative HPLC,and the separated compounds were identified by spectroscopic analysis.(4)The indexes are tested according to the four general rules of the Chinese Pharmacopoeia(2020 edition).To improve the character identification,powder identification,and thin-layer identification of the identification items;and to improve the moisture,ash,and acid-insoluble ash of the inspection items.The measurement items of the extract and the content determination item are increased.Results:(1)The original medicinal materials of Tibetan mu tong are Ranunculaceae plants,including Clematis montana Buch.-Ham.ex DC.,Clematis rehderiana Craib and Clematis brevicaudata DC.The medicinal parts are young branches with leaves and flowers and fruits about 2 to 5 mm in diameter.(2)The total extracts and total saponins of three basic plants of Tibetan mu tong had protective effects on gastric mucosa injury in rats,and Hederagenin had similar effects.The total saponins were better than the total extracts and the activity of Hederagenin was similar to that of the total saponins.In addition,the total saponins of each herb and Hederagenin could significantly reduce the expressions of TNF-α and IL-1β(P<0.01)in the serum of rats.All groups could reduce the content of MDA and SOD in the gastric tissue of rats,but the trend was not obvious.All groups could increase the epidermal growth factor EGF in gastric tissue.The NO content was significantly decreased(P<0.01).It is suggested that the protective effect of Tibetan mu tong on gastric mucosa is mainly related to antiinflammatory and immune regulation.(3)Ten compounds were isolated from the active fraction of Clematis brevicaudata DC.,including 3-(3-methoxy-4-O-β-Dglucopyranosylphenyl)Propan-1-ol(1)、clemastanin B(2)、Liriodendrin(3)、clematangoticoside F(4)、clematangoticoside B(5)、tanguticoside A(6)、saponin B(7)、clematangoticoside E(8)、Hederacoside C(9)、clematangoticoside D(10).(4)Development of draft quality standards for Tibetan mu tong,supplement character identification and powder identification under [identification],and supplement the thinlayer identification of Stigmasterol as a standard.Determine that the moisture content under [inspection] should not exceed 12%,and ash should not exceed 8%.To determine that the alcohol-soluble leachate under [extract] should not be less than 12.7%.It was determined that under [content determination],Hederagenin was used as an indicator component.This product was calculated as a dry product,and the content of Hederagenin in medicinal materials should not be less than 0.2%.Conclusion: In this paper,it was identified as the basic medicinal materials of Tibetan mu tong;the active fraction of Tibetan mu tong was identified as the total saponin parts.The chemical components of the active fraction of Clematis brevicaudata DC.were isolated and identified,and finally,10 compounds were obtained,all of which were isolated for the first time from this plant.Improved the existing quality standard of Tibetan mu tong. |
