Objective:In this study,the highly expressed gene TPM2 in liver cancer tissue screened by proteomics has been shown to play an important role in the progression of various tumors,but its role in hepatocellular carcinoma(HCC)has not been explored.Methods:The expression of TPM2 was verified by using TCGA,HCCDB and HPA databases and immunohistochemistry.Cox regression and Kaplan-Meier survival analysis were used to evaluate the prognostic value of.Go and KEGG enrichment analysis were also performed.The correlation between TPM2 and gene mutation was analyzed based on TCGA genome mutation data.ss GSEA,ESTIMATE algorithm and TISIDB and TISCH2 databases were used for tumor immunoanalysis.Immunotherapy efficacy and drug sensitivity were analyzed using TIDE score and R package "p RRophetic".Results:Through proteomic analysis and literature investigation,TPM2 was selected as the target of liver cancer in this study.The expression of TPM2 in different types of tumors is heterogeneous,but the expression level of TPM2 in liver cancer tissues is significantly higher than that in normal liver tissues.At the same time,TPM2 is related to the pathological grade and overall survival rate of liver cancer,and may promote the occurrence and development of liver cancer by affecting tumor metastasis and immunity.In addition,although TPM2 rarely mutates in liver cancer tissues,it is related to mutations of other genes,TMB,macrophage infiltration,immune regulation and tumor matrix components,and the group with high TPM2 expression is more prone to tumor immune rejection and drug sensitivity.Conclusion:As a highly expressed gene in liver cancer tissue,TPM2 may play a certain role in tumor metastasis,immune regulation and other aspects,and can be used as a potential target to predict the efficacy of immunotherapy and drugtherapy.
