| Objective:To investigate the impact of night shift work induced circadian rhythm disruption(CRD)on the expression of rhythm genes CLOCK in endometrial adenocarcinoma(EAC)and the possible mechanism involved in the progression of endometrial cancer.Methods:Selecteing the Fresh specimens of endometrioid adenocarcinoma that underwent surgical resection in our hospital from December 2021 to March 2022 and collecting the information of their previous night shift work history,according to background matching,endometrioid adenocarcinoma with a history of night shift work were selected as the abnormal rhythm group,and endometrioid adenocarcinoma without a history of night shift work were selected as the normal rhythm group.q PCR and Western blot were used to detect the expression of CLOCK m RNA and protein in the two groups.Paraffin tissues of32 patients with endometrioid adenocarcinoma who underwent surgical treatment in our hospital from August 2021 to December 2021 were collected.Immunohistochemistry was used in paraffin sections to detect the expressions of CLOCK protein and Epithelial mesenchymal transition(EMT)related phenotypic proteins E-cadherin and vimentin,and to analyze the relationship between the expressions of protein and clinicopathological features.Results:1.The relative expression levels of CLOCK m RNA and protein in EAC tissues of abnormal rhythm group were significantly lower than those of normal rhythm group(P <0.05);2.Compared with those without a history of night shift work,the positive expression rates of CLOCK and E-cadherin in EAC with a history of night shift work decreased,while the positive expression rates of vimentin increased,with statistically significant differences between the two groups,(P < 0.05);3.The positive expression rates of E-cadherin and CLOCK descended with the increase of FIGO stage and muscle infiltration depth,while the positive rate of vimentin increased,the difference between the groups was statistically significant,(P < 0.05).And there were no statistical significance in the expression of these proteins between different degrees of tissue differentiation,lymph node metastasis and age groups,(P < 0.05);4.The correlation between CLOCK expression and E-cadherin,vimentin in EAC tissue specimens was statistically different,(P < 0.05).Conclusion:1.The m RNA expression level and protein expression level of CLOCK gene in EAC with a history of night shift work were significantly lower than those in EAC without a history of night shift work,suggesting that CRD induced by night shift work is related to the differential expression of CLOCK gene.2.Compared with EAC without a history of night shift work,CLOCK positive expression rate decreased,E-cadherin negative expression rate and vimentin positive expression rate increased in EAC with a history of night shift work.The process of EMT promoted by CRD induced by night shift work is a possible mechanism involved in EC progression,which may be an important factor affecting prognosis.3.With the increase of FIGO stage and muscle infiltration depth,the positive expression rates of E-cadherin and CLOCK decreased,on the contrary,the expression of vimentin increased.This indicated that CLOCK expression and the incidence of EMT were related to the proliferation and invasion of endometrioid adenocarcinoma,and the specific mechanism needed to be further explored.4.The correlation between CLOCK expression and E-cadherin and vimentin in EAC tissue specimens was statistically different((P < 0.05)),indicating that CLOCK may be involved in EMT process,which requires further experimental verification. |
