Experimental Study On The Effect Of "Guoyanre" Acupuncture On MYOC And HSP72 In Primary Open-angle Glaucoma Rats

Objective:The main line of this study is the protective mechanism of "guoyanre" acupuncture to promote and regulate the retina.The main reference is the changes of intraocular pressure,retinal tissue morphology and central retinal artery blood flow index in primary open Angle glaucoma,and the main basis is the changes of related expression of MYOC,a key treatment factor,and HSP72,an important defense factor.Methods:Sixty SPF healthy SD rats(purchased from SPF Laboratory Animal Center of Gansu University of Traditional Chinese Medicine),half male and half female,6weeks of age,body weight 180-220 g,were selected as blank group by random number table,and the other rats were modeled.The model was made by injecting 0.3%compound carbomer solution into the anterior chamber pupillary area of both eyes for a total of 1 week.After the modeling,the intraocular pressure of the rats in the modeling group was measured,and 40 rats with good modeling quality were randomly divided into 4 groups: model group,supplementing and purging group,drug group and hyperocular fever group.The blank group was fed normally and the model was not replicated.The rats in the model group were bound and fixed during the intervention of the other groups.Relevant detection was carried out after the experiment.The model group was fed normally,the model was replicated,and the rats in the blank group were bound replication.After successful modeling in the Ping Bu Ping Xie group,the rats were placed in the rat fixator;Insert a fine needle into the "Fengchi point" of the rat for 0.5cm,and pull out the needle after even lifting,inserting and twisting;After successful modeling,the rats in the "guoyanre" group were placed in the fixator of rats,and a fine needle was inserted into the "Fengchi point" of the the needle was inserted,the indicator finger was pressed firmly at the point,and the thumb of the needle hand was rotated forward for 3-6 times.After feeling the needle sinking and tightening,continue to insert a small amount of heavy lifting times,so repeated operation 30 S,slowly out of the needle,after the needle with a cotton swab tightly press the acupoint 20 s,do not leave the needle;Once a day,5 days a course of treatment,3 courses of treatment.The intraocular pressure was measured before modeling and at the end of each course of treatment.The changes of blood flow in central retinal artery were detected before modeling and after each course of treatment.After all courses of treatment,the samples were taken and the retinal sections of rats were prepared.The data of changes in ocular pressure and blood flow,as well as the retinal m RNA and protein related data analyzed by fluorescence quantitative PCR,HE staining,immunohistochemistry and western blotting were collated and treated with spss26.0.Results:1.Changes of intraocular pressure in rats with primary open-angle glaucomaThe intraocular pressure increased in the model group(the trabecular reticulum space became smaller and the outflow of aqueous humor increased),the intraocular pressure improved in the drug group and the supplementing-pingxie group,and the improvement in the "guoyanre" group was the best.2.Effect on peak diastolic blood flow of CRA in rats with primary open-angle glaucomaCompared with blank group,blood flow diastolic peak value of CRA decreased in model group.After intervention,compared with model group,blood flow diastolic peak of CRA in drug group,Pingbupingpingpurgation group and "guoyanre" group was increased,and blood flow of CRA in "guoyanre" group was increased significantly.3.Morphological changes of retina in rats with primary open-angle glaucomaRetinal patching under light microscope of model rats showed thinning of the retina(cause: high intraocular pressure ischemia,resulting in retinal atrophy and degeneration,cell size reduction).Retinal patching under light microscope of the drug group and the supplementing and purging group showed increased retinal thickness,and the retinal thickness of the "guoyanre" group increased significantly.4.Effect on mrna expression of MYOC and HSP72 in primary open-angle glaucoma rats(1)The mrna expression of patients in the model group was increased compared with that of the blank group(P<0.05);After the intervention,compared with the model group,the m RNA expression of MYOC was decreased in the rats of the drug group,the supplementation and catharsis group and the "guoyanre" group,and the mrna expression of the patients of the "guoyanre" group was significantly reduced.(2)Compared with blank group,mrna expression of HSP72 in model group was increased(P<0.05);After intervention,compared with model group,the mrna expression of HSP72 was increased in drug group,Pingbu Pingpurgation group and "guoyanre" group(P<0.05).The mrna expression of HSP72 was significantly increased in the "guoyanre" group.5.Changes of MYOC and HSP72 protein expression in rats with primary open-angle glaucoma(1)Compared with blank group,the expression range of MYOC in model group was increased(P<0.05);After intervention,compared with model group,the expression range of MYOC in "guoyanre" group,Pingbu Pingxie group and drug group decreased(P<0.05).The expression range of MYOC decreased significantly in the "guoyanre" group(P<0.05).(2)Compared with blank group,the expression range of HSP72 in model group was increased(P<0.05);After intervention,compared with model group,the expression range of HSP72 in "guoyanre" group,Pingbu Pingxie group and drug group was increased(P<0.05).The expression range of HSP72 in "guoyanre" group was significantly increased(P<0.05).(3)Compared with blank group,MYOC expression in model group was increased(P<0.05);After intervention,compared with the model group,the expression level of MYOC in the drug group,the Pingbu Pingxie group and the "guoyanre" group decreased,and the "guoyanre" group had the most obvious decrease of MYOC expression.(4)Compared with blank group,the expression of HSP72 in model group was increased(P<0.05);After intervention,compared with model group,the expression level of HSP72 in drug group,Pingbu Pingxie group and "guoyanre" group was increased(P<0.05).The increase of HSP72 expression was the most obvious in the "guoyanre" group.Conclusions:"guoyanre" acupuncture in the treatment of primary open Angle glaucoma produces its mechanism by improving intraocular pressure,restoring retinal morphology,increasing blood flow in central retinal artery,and regulating the expression level of MYOC,a key pathogenic factor of POAG,and HSP72,a dual defense factor between retina and optic nerve.
