Objective Liposomes loaded with enolase 1 monoclonal antibody(ENO1 mAb-H1)were prepared to deliver ENO1 monoclonal antibody into target cells,and then the effect of ENO1 monoclonal antibody liposomes in cervical cancer Hela cells was studied.Methods 1.The optimal scheme for constructing liposomes was screened out by single-factor investigation by double emulsion solvent volatilization method,and then blank liposomes(Blank-Lips)and enolase 1 monoclonal antibody liposomes(ENO1mAb-H1-Lips)were prepared,and their physical characterization and morphological observation were made;2.Label liposomes with FITC and observe the uptake of ENO1 mAb-H1-Lips by cells by confocal microscopy;3.The cytotoxicity and proliferation inhibitory effects of Blank-Lips and drug-loaded liposomes on cervical cancer Hela cells were evaluated by CCK-8 experiment.Results 1.Successfully constructed drug-loaded liposomes,with a particle size of about 200 nm,spherical appearance,and basically the same size;2.The results of cell uptake experiments confirmed that Hela cells had a good effect on liposome uptake of monoclonal antibody coated with enolase 1,and the monoclonal antibody was successfully delivered into the cell.3.CCK-8 experimental results show that Blank-Lips has no toxic effect on Hela cells,Drug-loaded liposomes have no inhibitory effect on cell proliferation.Conclusion Liposomes successfully delivered ENO1 mAbs into cervical cancer Hela cells,and the preparation materials had no toxic effect on cells.
