| Objective:Female fertility preservation can be achieved by freezing embryos,oocytes,and ovarian tissue.Ovarian tissue cryopreservation and transplantation technology plays an important role in preserving female fertility and is an important way to solve the fertility problem of tumor patients.Studies have shown that after transplantation of frozen thawed ovarian tissue,ischemia/reperfusion and oxidative stress damage will lead to a large number of follicular necrosis or apoptosis,affecting the recovery of reproductive function after transplantation.The purpose of this study was to explore whether mitochondrial targeted antioxidant SS31 could reduce oxidative stress injury,reduce follicle apoptosis and improve the recovery of reproductive endocrine function after transplantation of rabbit frozen thawed ovarian tissue.Methods:The bilateral ovaries of rabbits were excised and made into fragments,which were cryopreserved by vitrification.One week later,the ovarian tissue was thawed and autotransplanted.SS31 was injected intraperitoneally after transplantation,NS was injected intraperitoneally in the control group,and the transplanted ovarian tissue was recovered on the 1st,3rd and 7th days after transplantation.The recovered ovarian tissue was stained with HE to observe the histological characteristics of the ovary,and the number of follicles at all levels and the number of normal follicles were counted;The MDA and SOD were detected by ELISA,and the oxidative stress reaction of frozen thawed ovarian tissue after transplantation was analyzed;Caspase3and Bax were detected to analyze the apoptosis of ovarian tissue after freezing and transplantation;The E2 and AMH were detected by ELISA,and the recovery of reproductive endocrine function was observed.Results: 1.The number of primordial follicles,total follicles and normal follicles in ovarian tissue of SS31 group was significantly higher than that of NS group on the 1st,3rd and 7th days after transplantation of rabbit frozen thawed ovarian tissue(P<0.05).2.On the 1st,3rd and 7th day after transplantation of rabbit frozen thawed ovarian tissue,the MDA content in SS31 group was significantly lower than that in NS group(P<0.05),while the SOD content in SS31 group was higher than that in NS group,with no significant difference.3.The content of Caspase3 and Bax protein in SS31 group was lower than that in NS group on the 1st,3rd and 7th day after transplantation of rabbit frozen thawed ovarian tissue,but the difference was not statistically significant.4.The AMH value and E2 value in SS31 group were higher than those in NS group on the 1st,3rd and 7th day after frozen thawed ovarian tissue transplantation in rabbits,and the difference was not statistically significant.Conclusions:After the transplantation of frozen thawed ovarian tissue,the number of original follicles,total follicles and normal follicles in the transplanted ovarian tissue were significantly increased after the application of mitochondrial targeted antioxidant SS31.SS31 may reduce follicle apoptosis by reducing the degree of lipid peroxidation and oxidative stress injury after ovarian tissue transplantation. |
