Objective: To detect the methylation level of histone H3K4 in gastric cancer tissue,and to reveal the relationship between the methylation level of histone H3K4 and clinicopathological features of gastric cancer.Methods: 1.A total of 263 cases of gastric cancer tissue and paired adjacent tissue specimens were selected from the Gastrointestinal Department of Guilin Medical College Affiliated Hospital from January 2015 to December 2018,with clear clinical diagnosis,no preoperative neoadjuvant treatment,and radical surgical resection.2.Apply the Epi Quik universal ubimethylated histone H3K4 quantitative analysis method to detect the methylation status of histone H3K4 in gastric cancer tissue and adjacent tissues.Use statistical methods such as chi square test or t-test to analyze the degree of methylation and its relationship with gender,age,tumor size,tumor location,tumor cell differentiation,clinical pathological staging,lymph node metastasis of gastric cancer The relationship between clinical and pathological features such as infiltration depth and distant metastasis.Results: The methylation level of histone H3K4 in gastric cancer tissue was significantly lower than that in adjacent tissues,with a statistically significant difference(P<0.05).2.The methylation level of histone H3K4 in gastric cancer tissue was not significantly correlated with the patient’s gender,age,tumor size,tumor location,and degree of differentiation(P>0.05);3.The degree of histone H3K4 methylation in gastric cancer tissue is correlated with the depth of tumor infiltration,lymph node metastasis,TNM staging,and distant metastasis,and the difference is statistically significant(P<0.05);4.TNM staging showed significant differences in H3K4 methylation levels between stages I and III,I and IV,and II and IV(P<0.05);However,there was no statistically significant difference in the degree of histone H3K4 methylation between groups in stages I and II,II and III,III and IV(P>0.05).Conclusion: 1.The methylation level of histone H3K4 in gastric cancer tissue is significantly lower than that in adjacent tissues,indicating that low methylation of histone H3K4 may be related to the occurrence and development of gastric cancer.2.The expression of histone H3K4 in gastric cancer tissue is closely related to the depth of tumor infiltration,lymph node metastasis,TNM staging,and distant metastasis in gastric cancer patients.This suggests that detecting the degree of histone H3K4 methylation can help determine the severity of the lesion in gastric cancer patients.
