Objective:1.Data mining was used to analyze medication rules and core drugs of digestive tract reactions after chemotherapy in the treatment of malignant tumors by the tutor,and to summarize the clinical experience of the tutor in the treatment of drugs;2.Network pharmacology was used to analyze the relationship between core drugs and digestive tract reactions after chemotherapy,and to analyze the mechanism of tutor therapeutic drugs.Methods:1.Collected the first clinical data and traditional Chinese medicine prescriptions of patients with digestive tract reactions after chemotherapy treated by Professor Renting-Li from October 01,2020 to October 01,2022 in the Oncology Department of the First Affiliated Hospital of Shaanxi University of Chinese Medicine and the outpatient department of famous medical library.After standardized data collation,Statistics and analysis were carried out by WPS Excel,SPSS Statistics 26.0,SPSS Modeler 18.0 and other software,including basic information,four Qi and five tastes of drugs,warp reduction efficacy,frequency of medication,association rule analysis,cluster analysis and network display.Thus,the medication rules and core drugs of the supervisor in the treatment of digestive tract reaction after chemotherapy were obtained,and the clinical experience of the supervisor in diagnosis,treatment and medication was summarized.2.According to the data mining results,the core drugs of digestive tract reaction after chemotherapy treated by the tutor were selected for network pharmacological analysis,and the active components of drugs were screened and corresponding target proteins were selected using TCMSP database;Disease gene targets were obtained from disease database,and disease-drug intersection targets were obtained,and intersection target Venny map was drawn.The intersection targets were imported into STRING database to construct PPI protein interaction network diagram.Cytoscape 3.9.1 software is used to construct Drug-Ingredient-Target-network diagram and drug-target-disease network diagram.The Metascape platform was used for KEGG and GO enrichment analysis,and bubble charts and bar charts were drawn to analyze the possible mechanism of tutor medication.Results:1.A total of 153 patients were included in this study,including 59 female patients(38.56%)and 94 male patients(61.44%).The proportion of male patients was higher than that of female patients,with a approximate ratio of 1.59:1;The minimum age was 30 years old,and the maximum age was 82 years old.The majority of patients were 60-69 years old(59 cases,38.56%)and 50-59 years old(48 cases,31.37%).Lung cancer(51 cases,33.33%),gastric cancer(26 cases,16.99%),and bowel cancer(27 cases,17.65%)were more common.2.A total of 173 traditional Chinese medicines were included in this study,and the four qi were mainly warm drugs(783 times,41.54%)and calming drugs(605 times,32.10%).Bitter drugs(1006 times,accounting for 33.53%),sweet drugs(913 times,accounting for 30.43%),octinic drugs(846 times,accounting for 28.20%)were the main five tastes.The splenic channel(1280 times,19.22%),lung channel(975 times,18.20%)and stomach channel(923 times,9.78%)were the main channels.The main effects were deficiency supplementing(520 times,accounting for 27.59%),humidification(232 times,accounting for 12.31%)and qi regulating(203 times,accounting for 10.77%).3.Among the drugs involved in this study,34 drugs were used for ≥20 times,the top5 were atractylodes atractylodes 126 times(5.68%),Magnolia officinalis 122 times(5.50%),Poria codonopsis 118 times(5.32%),Codonopsis codonopsis 106 times(4.77%),and Scorch malt 104 times(4.68%).Five common combinations were obtained by cluster analysis: C1: burnt malt,burnt Shenqu,Fructus aurantii,Magnolia officinalis,burnt hawthorn and Semen raphani;C2: amanita seed,Coix seed,atractylodes,poria cocos,codonopsis codonopsis,yam;C3: Peach kernel,safflower,Salvia miltiorrhiza,Zedoary,Scorpion;C4: Paeony,Angelica sinensis,raw land;C5: Platycodon platycodon,North Sea Ginseng,Ophiopogon ophiopogon,roasted licorice;Association rules were analyzed to obtain 30 groups of core drug pairs,including "Poria cocos → Atractylodes","Magnolia officinalis → Atractylodes","Codonopsis codonopsis → Poria cocos,atractylodes",and the correlation among drugs was shown through the network method.4.A total of 54 core drug active ingredients,480 targets,1006 disease targets,and170 interactional targets(12.9%)were obtained by network pharmacological analysis.The main active ingredients of the drugs were luteolin,honokiol,honokiol,poria acid,taraxyl alcohol,trichosanthol alcohol,phytosterol,etc.The main core targets were sarcoma gene(SRC),tumor suppressor gene TP53,signal transduction and transcriptional activation protein 3(STAT3),oncogene HSP90AA1,mitogen activated protein kinase 3 and 1(MAPK3,MAPK1),etc.GO enrichment analysis mainly involved biological processes such as positive regulation of protein phosphorylation,response to foreign biological stimuli,cell structures such as membrane rafts and cell bodies,and molecular functions such as protein kinase activity and tyrosine kinase binding proteins.KEGG enrichment analysis mainly involved cancer pathway,PI3K-Akt signaling pathway,AGE-RAGE signaling pathway,FoxO signaling pathway,HIF-1 signaling pathway,calcium signaling pathway and other signaling pathways.Conclusions:1.153 patients were included in this study,more males than females;Most of them are between 50 and 69 years old.Lung cancer,gastric cancer and bowel cancer are the most common diseases.2.173 TCM were involved in this study,and the four qi were mainly warm and flat;The five tastes are mainly bitter,sweet and sweet.The main channels are spleen,stomach and lung.The efficacy is based on supplementing deficiency,humidifying and regulating qi.3.The core drugs of tutor for digestive tract reaction after chemotherapy were:atractylodes atractylodes,Magnolia officinalis,poria codonopsis,and Char malt;And get 5common combinations.It shows that the teacher commonly used in the treatment of invigorating qi and strengthening the spleen,digestion and stomach,qi and water,promoting blood circulation and removing blood stasis,nourishing Yin and nourishing blood to comprehensive treatment,when necessary to add the products of dispelling evil and clearing collaments,detoxifying and dispersing,dispelling evil on the basis of fuzheng.4.The core drugs are characterized by multi-component,multi-target and multi-pathway treatment of digestive tract reaction after chemotherapy.The key effective components of drugs are mainly luteolin,magnolol and poria acid,and the key drug targets are SRC,TP53 and STAT3.The main pathways are cancer pathway,PI3K-Akt signaling pathway,AGE-RAGE signaling pathway,FoxO signaling pathway,etc.
