ObjectiveThe expression of SLP-2,ELK1 and Smad4 in papillary thyroid carcinoma(PTC)was investigated,followed by analyzing the relationship between the expression level of SLP-2,ELK1 and Smad4 in PTC as well as the relevance of the expression levels to clinicopathological characteristics of patients who had PTC,finally,the significance of SLP-2,ELK1 and Smad4 in the diagnosis and treatment of PTC was explored.Methods1.Cancer tissues and corresponding paracancer tissues of 30 patients with PTC(classical type)who met the inclusion criteria in the First Affiliated Hospital of Jiamusi University from January 2019 to August 2022 were collected,and paraffin-embedded tissues of 60 patients with PTC were collected(including the first 30 patients with tissue samples).2.The m RNA expression levels of SLP-2,ELK1 and Smad4 in caner tissues and corresponding adjacent tissues of PTC patient were detected by performing Real-time quantitative PCR.3.The protein expression of SLP-2,ELK1 and Smad4 in paraffin tissues of PTC patients were detected by immunohistochemical method.4.The relevance of m RNA and protein expression of the three factors to clinico-pathological characteristics of patients was investigated by using SPSS software.5.The relationship of the expression levels of three factors was detected by the usage of Spearman.6.The diagnostic performance of separate and combination applications of the three factors in papillary thyroid carcinoma was indicated by the area under ROC curve.Results1.Through q RT-PCR and immunohistochemistry,we founded that compared with paracancerous tissues,SLP-2 and ELK1 were significantly highly expressed in papillary thyroid carcinoma tissues;on the contray,Smad4 was prominently lower expressed in papillary thyroid carcinoma tissues than adjacent tissues.2.In the analysis of the expression and clinicopathological characteristics of patients,SLP-2,ELK1 and Smad4 expression levels of the three were correlated with TNM stage,lymph node metastasis and degree of tumor invasion(P<0.05).The molecular and protein expression levels of SLP-2 and Smad4 were correlated with the age of patients(P<0.05),in addition,the molecular expression levels of SLP-2 was also correlated with the tumor size(P<0.05).3.Spearman correlation analysis showed that SLP-2 was positively correlated with ELK1expression(r=0.662,P<0.001);Both expression were negative correlated with Smad4(r=-0.600,P<0.001 and r=-0.641,P<0.001).4.ROC curve showed that the area under ROC curve(AUC)of the combined detection of the three was 0.9833,the sensitivity was 100%,and the specificity was 90%,showing high diagnostic efficacy in papillary thyroid carcinoma.Conclusion1.SLP-2,ELK1 and Smad4 involve in the occurrence and development of papillary thyroid carcinoma.2.Overexpression of SLP-2 and ELK1 and low expression of Smad4 suggest poor prognosis.3.The combined detection of SLP-2,ELK1 and Smad4 is helpful to improve the diagnosis and prognosis of papillary thyroid carcinoma,and provide new directions and ideas for clinical treatment of papillary thyroid carcinoma.
