Objective: This study aims to investigate the chemical constituents from Patrinia villosa(Thunb.)Juss.and search for bioactive constituents and provide foundation of the quality standard and clinical application,which is advantageous to the development and utilization of P.villosa.The purpose of this study is also to provide components and compounds for the National Mega-Project for Innovative Drugs.Methods: The chemical constituents from whole plants of P.villosa were separated and puified by using a variety of modern chromatographic techniques,such as silica gel,Sephadex LH-20,MCI,ODS,MPLC,PHPLC,PTLC.The structures of isolated compounds were established by their spectroscopic analysis data of IR,HR-ESI-MS,NMR,CD and X-ray diffraction.Additionally,the anti-inflammatory effects of some isolated compounds inhibiting nitric oxide(NO)production in lipopolysaccharide-activated RAW 264.7 cells were assessed in vitro.Result: Fifty four compounds were isolated from the CH2Cl2 and Et OAc partitions of P.villosa,including 11 iridoids,four ionone compounds,seven triterpenoids,seven lignans,9 simple phenylpropanoids,2 coumarins,5 steroid,2 flavonoids,5 others.The absolute configuration of B1,B2,B5,B6 were determined by CD,whereas that of 3was based on X-ray diffraction.The anti-inflammatory activities of 16 iridoids and triterpenoids were screened.Among them,compounds B1,B2,B5,B6,B9,B16,B17,B18,B20,B21,B22 exhibited anti-inflammatory with IC50 values of 18.14 μM,18.93μM,22.00 μM,13.64 μM,26.41 μM,47.15 μM,31.14 μM,25.65 μM,25.75 μM,16.49μM,25.65 μM.The IC50 values of compounds B3,B4,B10,B11,B19 are more than100 μM.Conclusion: Among the 54 compounds,9 compounds(B1-B6,B22,B33,B34)are novel compounds and 38 known compounds are isolated from P.villosa for the first time.Compounds B1,B2,B5,B6,B9,B17,B18,B20,B21,B22 exhibited strong anti-inflammatory and compounds B16 showed medium anti-inflammatory.
