the effect of butyric acid on the rumen function of goats fed with high-concentration diets, involving rumen fermentation parameters, rumen tissue morphology, lipopolysaccharide and blood inflammatory factors,as well as Metabolome and other aspects. The experiment is divided into three parts.Part 1: The effect of different levels of butyric acid on the rumen of goats fed with high-concentration feed.In this experiment,24 Dazu black goats(body weight 14.2±0.4kg)equipped with permanent rumen fistula were randomly divided into 4 groups, namely control group(CON),low butyric acid group(LD,0.3g/kg body weight),Medium butyric acid group(MD,0.5g/kg body weight)and high butyric acid group(HD,0.7g/kg body weight). The results showed that the MD and HD groups significantly reduced the feed intake of goats.Different levels of butyric acid did not reduce the rumen p H of goats fed with high-concentration diets to below 5.8,and did not cause SARA in goats. The LD group significantly reduced the concentrations of acetic acid, propionic acid, butyric acid and total volatile fatty acids;the MD group significantly reduced the concentrations of acetic acid and propionic acid, and the concentration of butyric acid and total volatile fatty acids;the HD group significantly reduced the concentrations of acetic acid,Concentrations of propionic acid, butyric acid and volatile fatty acids. The LD group significantly reduced the trypsin activity, and the MD and HD groups significantly reduced the trypsin activity. The MD and HD groups significantly reduced the concentration of lactic acid in the rumen fluid, and the LD group significantly reduced the concentration of lactic acid in the rumen fluid on the 28 th and 35 th days.On the 28 th day,the rumen fluid LPS of the LD group was significantly lower than that of the CON and HD groups. The blood LPS of the LD group was extremely significantly lower than that of the HD group, and significantly lower than that of the MD group. On the 35 th day,the rumen fluid LPS of the LD group was significantly lower than that of the HD group, and the blood LPS of the LD group was significantly lower than that of the MD and HD groups. On days 7,14 and35,the HD group significantly increased the alkaline phosphatase concentration, and on the 28 th day,the HD group significantly increased the alkaline phosphatase concentration. The LD group significantly reduced the concentration of interleukin-6;the HD group significantly reduced the concentration of immunoglobulin G,insulin, and glucose, and significantly reduced the concentration of immunoglobulin M;the MD group significantly reduced immunoglobulin G and immunoglobulin M concentration. The above results indicate that with the increase of butyric acid levels, the rumen fermentation of the high-concentrate fed goat diet is inhibited, which reduces the immune capacity of the high-concentrate fed goats, and does not cause the goats to develop SARA.Part 2: The effect of butyric acid on the rumen of goats fed with different concentrate to forage ratios. In this experiment,24 Dazu black goats(body weight 15.6±0.6kg)equipped with permanent rumen fistulas were selected and randomly divided into 4 groups, namely, the high-precision control group(HC-CON)and the high-precision butyric acid group(HC-BUT,0.4g/kg body weight),high coarse material control group(HF-CON)and high coarse material butyric acid group(HF-BUT,0.4 g/kg body weight). The results showed that butyric acid significantly reduced the feed intake of goats fed with high-concentration feed.Butyric acid did not cause the p H of the rumen of goats fed with high-concentration and high-coarse materials to drop below 5.6,and did not cause SARA in goats.Butyric acid significantly reduces the concentration of acetic acid in the rumen fluid of goats fed with high-precision feed, and significantly reduces the concentration of propionic acid and butyric acid.Butyric acid significantly improves the activity of goat chymotrypsin fed with high coarse feed.Butyric acid significantly reduces the concentration of lactic acid in the rumen of goats fed with high-precision feed, and significantly reduces the concentration of blood lactic acid.Butyric acid significantly reduced the concentration of LPS in the rumen fluid of goats fed with high coarse feed.Butyric acid significantly increased the concentration of total bilirubin in goats fed with high-concentration feed.Butyric acid significantly reduced the concentration of tumor necrosis factor-α in goats fed with high-concentration feed.Butyric acid significantly reduces the length of the rumen papilla of goats fed with high-concentration feed, and significantly reduces the width of the stratum corneum. The above results indicate that butyric acid has no significant effect on the rumen fermentation of goat diets fed with high coarse feed, which is different from that butyric acid has a strong inhibitory effect on rumen fermentation of goat diets fed with high concentration feed.Butyric acid has no significant effect on the epithelial morphology and immune factors of the high-coarse diet. The addition of butyric acid is not conducive to the immune function of goats fed with high-concentration diets.Part 3: the effect of butyric acid on the rumen fluid and blood metabolome of goats fed with different concentrate to forage ratios. In this experiment, butyric acid(0.4g/kg body weight)significantly up-regulated the differential metabolite acetylcholine in the rumen of goats fed with high concentrate. On the one hand,it promotes the conversion of acetylcholine into acetic acid under the action of acetylcholinesterase, and on the other hand promotes the effect of vesicular acetylcholine transporter on the The transport efficiency of acetylcholine promotes the normal function of cholinergic synaptic metabolism pathway.Significantly down-regulate the differential metabolite adenylate in the rumen and inhibit the normal function of the c AMP signaling pathway;significantly down-regulate the differential metabolite uracil in the rumen and inhibit the β-alanine metabolic pathway.Butyric acid perfusion promotes the synthesis of arachidonic acid metabolic pathway in the rumen of goats fed with high coarse feed.Butyric acid perfusion high-precision feed in goats inhibits the normal function of pyrimidine metabolic pathways by up-regulating cytidine in the blood;up-regulating docosahexaenoic acid in the blood and promoting the biosynthesis of unsaturated fatty acids.Feeding goats with butyrate perfusion high coarse feed significantly down-regulated ascorbic acid in the blood and inhibited the normal function of HIF-1signaling pathway;by significantly up-regulating D-galactosamine in blood,it promoted the synthesis of galactose metabolism pathway. The above results show that the effect of butyric acid on the amount of metabolites in the rumen fluid of goats fed with high coarse feed is lower than that of the high concentrate group, but the changes in blood metabolites are not much different. | | Keywords/Search Tags: | butyric acid, rumen fermentation, inflammatory factors,
