| Rice stripe disease,caused by rice stripe virus(RSV),is an important virus disease in rice production.p2,encoded by RSV,is a nucleoplasmic shuttle protein.It interacts with nucleolar protein fibrillan2(Fib2)and cajal body protein coilin in the cell nucleus,involved in virus replication and movement.In addition,p2 can interact with gene silencing repressor 3(SGS3)in the cytoplasm.SGS3,a cofactor of RNA-dependent RNA polymerase 6(RDR6),combines with RDR6 as small interfering RNA(si RNA)bodies.SGS3-RDR6 bodies play an important role in plant disease resistance and immunity mediated by RNA silencing.The relationship of p2 and SGS3-RDR6 bodies were identified in this thesis,the results laid a foundation for p2 biological function.The thesis contains two parts as follows:1.Interaction study between p2 and Nb SGS3-RDR6 bodies: the relationship between p2 and Nb SGS3-RDR6 bodies was identified by subcellular co-localization and Bimolecular fluorescence complementation(Bi FC).2.The preliminary investigation of the interaction mechanism between p2 and Nb SGS3-RDR6 bodies: transient expression of p2,SGS3,and RDR6,western blot showed that p2 mediated Nb SGS3 degradation,but failed to affect the expression of Nb RDR6.After treatment with the proteasome inhibitor MG132 and the autophagy inhibitor 3-MA,it was found that p2-mediated degradation of SGS3 may be generated by the ubiquitin-proteasome system and the autophagy pathwayIn summary,this thesis revealed that p2 interacted with Nb SGS3-RDR6 bodies and might degrade SGS3 expression through the ubiquitin-proteasome system and autophagy pathway.Those results lay a foundation for p2 biological function. |
