ObjectiveThis research observed the effect of Tongmai Yishen Formula on the expression of ADM and E-cadherin protein in kidney of rats with unilateral ureteral obstruction,to explore the antagonism of renal interstitial fibrosis and possible mechanism and provide new ideas for clinical prevention and treatment and drug screening of renal interstitial fibrosis.Method50 male SD rats were randomly divided into 5 groups:sham-operated group,model group,Tongmai Yishen Formula group,valsartan group,ADM treatment group.There are 10 rats in each group,and all the rats were gavaged respectively with normal saline,normal saline,Tongmai Yishen Formula,valsartan,normal saline and ADM mini-Osmotic pump by subcutaneous administration for treatment of 14 days.The content of ADM in kidney and plasma was estimated after each group was sacrificed;after removing the diseased side kidney,Masson staining was made after paraffin section to observe the pathological morphological changes of the renal tissues;E-cadherin protein detected by western blotting.Results1.In model group,there existed a large number of inflammatory cells infiltration in renal interstitial tissue,fibrous tissue hyperplasia,decreased number of glomerulus,and partial expansion,atrophy and lumen collapse in renal tubules.The renal interstitial lesion in the rats of Tongmai Yishen Formula Group was significantly lower than that of the model group.2.Compared with model group,the expression of ADM and E-cadherin protein in the kidney tissues of each treatment group was increased(P<0.01),and there was no significant difference between 22 groups(P>0.05).3.Compared with model group,the expression plasma ADM protein was decreased in Tongmai Yishen Formula group and valsartan group(P<0.01),there was no significant difference between the two treatment groups(P>0.05);the expression of ADM in shamoperated group and treatment group of ADM protein in plasma were higher than that in model group(P<0.01),while no statistical significance between the two(P>0.05).4.The content of ADM protein in renal tissue was positively correlated with the expression of E-cadherin protein,and the content of E-cadherin protein increased with the increase of ADM content.(r=0.44,P<0.01)Conclusion1.Tongmai Yishen Formula showed the ability to reduce the inflammatory cell infiltration,the proliferation of fibrous tissue,and improve the renal interstitial fibrosis caused by unilateral ureteral ligation.2.Tongmai Yishen Formula up-regulated the expression of ADM in kidney tissue and Ecadherin protein,and showed a positive correlation between the two.We can infer that the protective function of Tongmai Yishen Formula may work through up-regulation of local ADM protein content in kidney regardless of circulating ADM and the antagonism of the transdifferentiation of renal tubular epithelial cells.This has the potential to be one of the mechanisms of renal interstitial fibrosis.
