| Objective: To observe the effect of the pressing manipulation on the expression levels of 5-HT2 B receptor protein and 5-HT2 C receptor protein and the changes of GABA content in the spinal cord of rats with cer ebral apoplexy muscle spasm.Methods: A random number table was used to divide 24 rats into 4 groups,including blank group,sham operation group,model group and treatment group,respectively.The model group and treatment group constructed the MCAO cerebral ischemia model,and a same operation was performed on the sham operation group but without inser ting the suture.Afterwards,the muscle tension level of the rats was measured by the modified Ashworth method,the motor function of the rats was measured by the balance beam experiment,the expression of 5-HT2 B receptor protein and 5-HT2 C receptor protein in the spinal cord was measured by Western-Blot,and the GABA content in the spinal cord was measured by ELISA.Results: The improved Ashworth muscle tone score shows that the muscle tone of model group and treatment group were significantly higher than the blank group and sham operation group(P<0.05),and the treatment group was significantly lower than the model group(P<0.05);The balance beam exercise ability score shows that the exercise ability of blank group and sham operation group were significantly higher than the treatment group and model group(P<0.05),and the exercise ability of model group was significantly lower than the treatment group(P<0.05);The Western-blot shows that the amount of5-HT2 B and 5-HT2 C in model group and treatment grou p were significantly higher than the sham operation group and blank group(P<0.05),and the amount of 5-HT2 B and 5-HT2 C in model group was higher than the tre atment group(P<0.05).The ELISA result shows that the amount of GABA in sham operation group and blank group were higher than model group and trea tment group(P<0.05),and the GABA in treatment group is higher than model group(P<0.05).Conclusion: After treatment,the muscle spasm of rats after stroke can be effectively relieved,muscle tension can be r educed,and the recovery speed of motor ability can be improved.Its effect mechanism may be achieved by inhibiting the expression of 5-HT2 B receptors and 5-HT2 C receptors in the spinal cord,thereby increasing the release of GABA. |
