Purpose: Based on data mining method,study and summarize the medication rule of tutor in treating post-infectious cough,explore the mechanism of action of core prescription in treating post-infectious cough by using network pharmacology,and provide basis for TCM treatment of post-infectious cough.Materials and METHODS: The relevant data of patients with post-infectious cough treated by our tutor in the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from May 1,2020 to December 30,2021 were collected.A total of 382 medical records were included according to the criteria of inclusion and classification,and relevant data were extracted to establish a database.Excel,SPSS Modeler 18.0 and SPSS Statistics 25 were used for descriptive statistical analysis,association rules and cluster analysis of prescription data.Based on the above analysis results combined with clinical practice,the academic thoughts and core prescriptions of tutor’s post-infection cough were further summarized.The effective ingredients and target information of core prescriptions were screened by TCMSP and Batman-TCM database,and the gene names corresponding to target were obtained by Uniprot database.Gene targets of post-infection cough were obtained by Gene Cards and Dis Ge NET disease gene databases.Venny2.1 software was used to draw Venn diagram to obtain drug disease co-action targets.Using Cytoscape3.9.0 software to draw a network diagram of "TCM-component-target";Protein interaction network graph was constructed by String database.DAVID and KOBAS databases were used for enrichment analysis of action targets,so as to explore the mechanism of action of core prescription in treating post-infection cough.Conclusion:1.Syndrome Type: Among the 382 patients,the syndrome with the highest frequency was phlegm-heat closed lung syndrome(20.7%).The second was the syndrome of cold attack on lung(19.1%),phlegm and dampness accumulation of lung(14.1%),deficiency of lung qi(13.4%),lung heat and Yin deficiency(12.6%),wind-heat invasion of lung(10.7%),wind-dryness injury to lung(5.8%),liver qi stagnation(3.7%).2.Medication Status: The 382 prescriptions contained 105 TCM ingredients,of which 25 were more than 20% in frequency.According to the highest frequency of use,the order was Platycodon platycodon,prepared glycyrrhiza,Tangerine peel,bitter almond,Anthurium,Aster,Schizonepeta schizonepeta,Baibu,Atractylodes atractylodes,Astragalus membranaceus,Radix astragalus officinalis,Radix Ophiopogonis,Fructus aurantii,Radix Bupleurum,Loquat leaves,radix scrophulariae,Ligusticum chuanxiong,Fritillaria thunbergii,Angelica,Poria cocos,Xanthii fructus,Scutellariae baicalensis,Radix Cynanchii and Radix Rehmanniae.Warm,cold,flat drugs are used more;The medicinal taste is mainly bitter,spicy and sweet.The return meridian is mainly lung meridian,spleen meridian,stomach meridian,liver meridian and heart meridian.Drugs such as relieving phlegm,relieving cough and relieving asthma,tonifying deficiency,relieving surface and clearing heat were frequently used.3.Cluster analysis: the drugs could be grouped into 6 categories.The first category was:Herba,Aster,Bitter almond,Schizonepeta schizonepeta,Baibu,Tangerine peel,loquat leaf and Platycodon grandiflorum.Type 2: Radix Ophiopogonis,Radix Aristophanae,Radix scrophulariae,Radix Rehmanniae,Fructus aurantii,Radix Bupleurum,Radix Scutellariae;The third category : Radix Cyrrhizae,Xanthii fruit,Astragalus membranaceus,Atractylodes macrocephala,parsnip;Category 4 : Ligusticum chuanxiong,Angelica sinensis;Class 5: Poria cocos,Pinellia ternata;Class 6:processed licorice4.Association rules: Common drugs for Radix Cyrrhizae and Xanthii fructus;Radix Ophiopogonis,Radix Ophiopogonis;Bupleurum and fructus aurantii;Aster.Combined with the clinical experience of the tutor,the core prescriptions were made as follows:Rhizome,aster,bitter almond,Schizonepeta,Baibu,Tangerine peel,loquat leaf and Platycodon grandiflorum.5.There were 88 active ingredients in the core prescription,among which quercetin,luteolin,β-sitosterol,galate and kaempferol were the main active ingredients.There were130 potential targets corresponding to drugs and diseases,mainly STAT3,JUN,MAPK3,MAPK1,MYC and so on.GO enrichment analysis of biological processes mainly involved cytokine mediated signaling pathways,positive regulation of gene expression,estradiol response,positive regulation of RNA polymerase II promoter transcription,lipolysaccharides response,hypoxia response,aging,etc.KEGG pathway enrichment analysis mainly involved age-rage signaling pathway and TNF signaling pathway in diabetic complications.Conclusion:1.My tutor believed that external evil invaded the lung,and the loss of lung xuan was its basic pathogenesis,which was closely related to the pathogenic factors of "wind","phlegm" and "deficiency".The disease was located in the lung and related to the spleen,stomach and liver organs.The drug used in the supervisor’s prescription is mainly warm,cold and flat;the drug tastes are mainly bitter,spicy and sweet;the return channels are mainly lung channels,spleen channels,stomach channels and liver channels;the drug characteristics are one xuan and one fall,one clear and one run,one rise and one fall,one line and one powder,etc.;the treatment is mainly to xuan and moisten the lung,reduce the lung and tonify the lung.He put forward the theories of "seeking the root of cure","treating lung without cough","treating phlegm first" and so on.2.Quercetin,luteolin,β-sitosterol,galate,kaempferol and other active components contained in the core prescription pass through age-rage signaling pathway in diabetic complications and TNF signaling Pathway regulates STAT3,JUN,MAPK3,MAPK1,MYC and other targets,which can stimulate the generation of inflammatory factors secreted by monocytes,activate the transcriptional activity mediated by antioxidant stress response elements,inhibit the expression of inflammatory factors,and thus reduce airway inflammation and airway secretion.
