Purpose : To observe the effects of traditional Chinese medicine HEWei Fanliukang on the expression of Wnt3 a and β-catenin in chronic atrophic gastritis rats,to prove the possible theory of their treatment of chronic atrophic gastritis,And provide new views and methods for the treatment of chronic atrophic gastritis Materials and methods: Forty SPF Wistar rats with body weight of 200±20g were randomly divided into two groups:model group(n = 32)and blank group(n = 8).The model group was given intragastric fluid.The model was intragastric with the prepared perfusion fluid,150g/L,55℃ hot saline,10 ml · kg-1 · d-1,and the model was intragastric.2% salicylic acid solution 10 m L · kg-1 · d-1,gavage;All were once /d;20mmol/L deoxycholate sodium solution,free to drink;Hunger and satiety disorder treatment,namely 2d satiety + 1D fasting.modeling 16 weeks.Successful modeling rats were indiscriminately divided into 3 groups,Including model group,western medicine group,traditional Chinese medicine group,with each group of nine only.The rats in the blank group and the model group were given the same amount of distilled water,the rats in the western medicine group were given 18.75 mg/kg body weight,and the rats in the traditional Chinese medicine group were given 27.3 g/kg body weight,each administration group after the end of self-modeling continuous administration,once a day.During the treatment,the general condition of the rats was closely observed.After treatment,all rats were fasted without water for 24 h.After anesthesia,the abdominal cavity was opened and gastric specimens were taken.Through the macroscopic observation,the gastric tissue was trimmed,fixed and paraffin embedded.HE staining and light microscopy were used to observe the pathological changes of gastric mucosa tissues of rats in each team.Western-blot method was used to detect the expression levels of Wnt3 A protein and β-catenin protein in rat gastric tissue;and the m RNA levels of WNT3 A andβ-catenin(CTNNB1)in gastric tissues were spotted by RT-q PCR method.Results:1.During modeling,it was observed that rats with chronic atrophic gastritis had dark hair,irritability in the early stage,drowsiness and weight loss after modeling.After treatment,the body weight of mice increased obviously,the fur was shiny,and the mental state was improved obviously.2.Aatrophy of gastric glands or inflammatory reaction,white gastric mucosa and flattening or disappearance of gastric mucosa were observed by naked eyes.HE staining showed that gastric mucosa glands in the model group were atrophic and decreased in number,buffer layer thickness and inflammatory cell infiltration.After administration,compared with model group,gastric mucosa atrophied in rats.Rats in each treatment group had obvious improvement,body number increased,more orderly arrangement,gastric mucosa mucosa increased,inflammatory cells decreased.In particular,the effect of the Korean medicine group was the best,which was closest to that of the commonly used medicine group.3.Western-blot test results: Compared with the white group,the expressions of Wnt3 a protein and β-catenin in the gastric tissue of rats in the model group were expressively increased,and the expressively was statistically significant(P < 0.05).Compared with the model group,the expressions of Wnt3 a protein and methylcatenin in gastric tissues of Western medicine group and Chinese medicine group were significantly reduced,and the difference was statistically significant(P < 0.05).Compared with the western medicine group,the expression of Wnt3 a protein and glutamine in gastric tissue of rats did not show any significant difference.(P>0.05).4.RT-q PCR detection results: RT-q PCR detection of Wnt3 a and CTNNB1 gene expression in rat gastric tissue showed that:(1)Compared with blank group,the expression levels of Wnt3 a and CTNNB1 gene in gastric tissue of model group were significantly increased(P < 0.05);(2)Compared with the model group,the expression levels of Wnt3 a and CTNNB1 in the gastric tissue of the Chinese medicine group and the western medicine group were both decreased(P<0.05);(3)There was no significant difference between the Chinese medicine group and the western medicine group(P>0.05).Conclusion:1.Treatment of CAG with Gastric reflux kang can improve the atrophy of gastric mucosa.2.The mechanism of HEWei Fanliukang prescription improving gastritis mucosa atrophy of CAG may be related to regulating the expression of Wnt3 a protein andβ-catenin protein.
