Objective: To analyze and summarize the medication rules of Traditional Chinese Medicine(TCM)for the treatment of ischemic cardiomyopathy using data mining techniques,providing theoretical and practical basis for the diagnosis and treatment of this clinical disease.Furthermore,network pharmacology methods are used to explore the active ingredients,targets,and mechanisms of the strongest drug associations in TCM treatment of ischemic cardiomyopathy.Materials and Methods:1.Chinese medicine formulas and patent medicines used for treating ischemic cardiomyopathy were collected from databases including CNKI,Wanfang,VIP,Pub Med,and the Outpatient Department of Liaoning University of Traditional Chinese Medicine.Screening was performed based on the inclusion and exclusion criteria,and the results were standardized according to uniform standards.Finally,the collected Chinese medicine information was transformed into data format and entered into a database,and software such as WPS Excel and the Traditional Chinese Medicine Inheritance Calculation Platform V3.0were used for statistical analysis.The medication rules for treating ischemic cardiomyopathy with Chinese medicine were discussed and summarized.2.Based on network pharmacology,the pharmacological mechanisms of the strongest associated drug pair(Danshen-Honghua)used in traditional Chinese medicine to treat ischemic cardiomyopathy were analyzed.The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was used to retrieve the effective components and related targets of Danshen and Honghua,and the "drug-active ingredient-key targets" network was constructed using Cytoscape software.The PPI network was generated from the STRING platform,and GO analysis and KEGG pathway enrichment analysis were performed in the Metascape database.Finally,the pharmacological mechanisms of the strongest associated drug pair for treating ischemic cardiomyopathy were predicted.Results:1.A total of 95 cases,including 75 outpatient medical records and 20 literature cases,were finally included in this study.Among them,there were 22 males and 73 females,and the highest incidence of the disease was in the 60-69 age group,with the predominant pattern being Qi deficiency and Blood stasis.There were 219 prescription records involving 199 Chinese herbs,and the cumulative frequency of medication use was 3750 times.When the frequency of use was ≥ 90 times,it was classified as a high-frequency medication.The commonly used effects were promoting blood circulation and removing blood stasis,nourishing deficiency,calming the mind,and relieving exterior syndrome.The main medicinal properties were warm,neutral,and cold,with the main flavors being sweet,bitter,and pungent,and the main channels being the liver,heart,and spleen.Association rule analysis showed that Danshen-Honghua was the strongest associated drug pair,and the most frequently used herbal formula was composed of Taoren,Danshen,and Honghua.Through the K-means algorithm and regression clustering model,three core combinations were obtained: Combination 1: Danshen,Honghua,Danggui,Taoren,Huangqi;Combination 2:Taoren,Huangqi,Chuanxiong,Danshen,Taizishen;Combination 3: Taoren,Honghua,Danggui,Danshen,Baizhu.2.Using network pharmacology methods,a chemical-target network for Danshen-Honghua in the treatment of ischemic cardiomyopathy was obtained,which included 76 compounds and 267 corresponding targets,with 103 drug intersection targets.The active ingredients of the drugs mainly included quercetin,luteolin,kaempferol,salvianolic acid A,and baicalin.The key targets in the protein-protein interaction network were AKT1,IL6,TNF,VEGFA,TP53,CASP3,and IL1 B.The effective pathways for affecting ischemic cardiomyopathy were found to be Pathways in Cancer,AGE-RAGE Signaling Pathway in Diabetic Complications,TNF Signaling Pathway,etc.Conclusion:1.Data mining results: The treatment of ischemic cardiomyopathy in traditional Chinese medicine(TCM)mainly uses herbs with sweet,bitter,and pungent flavors,which primarily enter the liver,heart,and spleen meridians.The core herbs include peach kernel,Chinese angelica,safflower,salvia miltiorrhiza,astragalus,chuanxiong,white atractylodes,and prince ginseng.The most commonly used herb pairs are salvia miltiorrhiza-safflower,peach kernel-safflower,and salvia miltiorrhiza-peach kernel,and the most commonly used herb trio is salvia miltiorrhiza-safflower-peach kernel.Three frequently used TCM formulas for the treatment of ischemic cardiomyopathy were obtained through cluster analysis.2.Network pharmacology results: The pharmacological mechanism of the most strongly associated herb pair,salvia miltiorrhiza-safflower,was analyzed.The active components of salvia miltiorrhiza-safflower were found to be mainly composed of quercetin,luteoloside,kaempferol,tanshinone IIA,and baicalein.A total of 103 common targets were obtained,and the top seven effective targets were AKT1,IL6,TNF,VEGFA,TP53,CASP3,and IL1 B.GO analysis mainly enriched several biological processes such as kinase binding,DNA binding transcription factor binding,and protein domain-specific binding.KEGG pathway analysis showed that the salvia miltiorrhiza-safflower herb pair may affect pathways related to cancer,the AGE-RAGE signaling pathway in diabetic complications,the TNF signaling pathway,and several others.
