| Objective: By observing the effect of Zhuluan decoction on the rat model of Premature ovarian insufficiency induced by cyclophosphamide,the protective mechanism of Zhuluan decoction on the decline of early onset ovarian reserve function in rats was explored by enzyme-linked immunosorbent assay and protein blotting,which is helpful to guide the clinical application of Zhuluan decoction to improve symptoms.Methods: Sixty clean female SD rats were randomly divided into normal group(10 rats)and model group(50 rats).The model group was injected intraperitoneally with cyclophosphamide(CTX)according to the specifications for 15 days to induce the rat model of Premature ovarian insufficiency.After the evaluation of the success of the model,the model group was divided into model group,progynova group and high,medium and low dose groups of Zhuluan Decoction(10 rats)according to the principle of random grouping.Normal group and model group were gavaged with normal saline,progynova group was gavaged with 01.mg/kg,and the high,medium and low dose groups of Zhuluan decoction were gavaged with56,28,14g/kg.The rats were killed after 21 days of continuous administration.The estrous cycle and body mass of rats in each group were detected.The content of serum hormone(FSH,LH,AMH)in rats was detected by enzyme-linked immunosorbent assay(ELISA),and the expression of LC3 protein in rats’ ovaries was detected by Western blot.Results: Compared with the normal group,the estrous cycle in the model group was prolonged,stagnant or even disturbed.Compared with the normal group,the body mass of the model group was decreased(P < 0.01),and the body mass value of all groups after medication was increased(P < 0.01,P <0.05).Compared with normal group,the serum values of folliclestimulating hormone(FSH)and luteinizing hormone(LH)in model group were increased(P < 0.01),while the serum values of follicle-stimulating hormone(FSH)and luteinizing hormone(LH)in Bujiale group and Chinese medicine group were decreased compared with model group(P < 0.01,P <0.001).Compared with the normal group,the level of serum anti-mullerian hormone(AMH)in the model group decreased(P < 0.01).Compared with the model group,the level of serum anti-mullerian hormone(AMH)increased(p< 0.01),and the expression of LC3 protein in the model group increased(p< 0.01),the expression level in each group was decreased(p < 0.01,p <0.05).Conclusion: Zhuluan Decoction can decrease the level of serum Follitropin beta(FSH)and Lutropin Alfa(LH),and increase the level of serum anti-mullerian hormone(AMH)to improve the ovarian reserve function and inhibit the occurrence of premature ovarian insufficiency It can decrease the expression of LC3 protein in POI rats and improve the ovarian function. |
